Activity Data (IL17 (interleukin 17) is a member of IL17 cytokine family, which is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. It is reported that IL17 is a ligand for IL17RA, indicating its interaction with IL17RA. Thus, a binding ELISA assay was constructed to detect the association of recombinant human IL17 with recombinant human IL17RA. Briefly, IL17 were diluted serially in PBS with 0.1%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17RA-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution , wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL17 with IL17RA was shown in Figure 1 and this effect was in a dose dependent manner.)

Activity Data (Figure. Cell proliferation of ECV-304 cells after stimulated with VEGFA.Vascular endothelial growth factor A (VEGFA) is a protein that in humans is encoded by the VEGFA gene. This protein is a glycosylated mitogen that specifically acts on endothelial cells and has various effects, including mediating increased vascular permeability, inducing angiogenesis, vasculogenesis and endothelial cell growth, promoting cell migration, and inhibiting apoptosis. VEGFA shows prominent activity with vascular endothelial cells, primarily through its interactions with the VEGFR1 and -R2 receptors found in prominently on the endothelial cell membrane. Thus, proliferation assay of recombinant human VEGFA was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640  prior to the addition of various concentrations of VEGFA. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of ECV-304 cells after incubation with VEGFA for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with human recombinant VEGFA for 48h. The result was shown in Figure 2. Soluble CX3CL1 potently chemoattracts T cells and monocytes, while the cell-bound chemokine promotes strong adhesion of leukocytes to activated endothelial cells, where it is primarily expressed.Thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of CX3CL1 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (150uL cell suspension, 106 cells/mL in RPMI 1640 with FBS free) and SLC (15.625ng/mL, 31.25ng/mL, 62.5ng/mL and 125ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. Result shows CX3CL1 is able to induce migration of THP-1 cells. The migrated Jurkat cells in low chamber at low magnification (×100) were shown in Figure 1. Five fields of each chamber were randomly chosen, and the migrated cells were counted at high magnification (×400). Statistical results were shown in Figure 2. Activity Data (IL20 (Interleukin-20) is a cytokine structurally related to interleukin 10, which is produced by activated keratinocytes and monocytes. Activity Data (Figure. The binding activity of SEMA3A with NRP1.Semaphorin 3A (SEMA3A) is a member of the semaphorin family. Semaphorin 3A is secreted by neurons and surrounding tissue to guide migrating cells and axons in the developing nervous system. This secreted Sema3A protein can function as either a chemorepulsive agent, inhibiting axonal outgrowth, or as a chemoattractive agent, stimulating the growth of apical dendrites. In both cases, the protein is vital for normal neuronal pattern development. Besides, Neuropilin 1 (NRP1) has been identified as an interactor of SEMA3A, thus a binding ELISA assay was conducted to detect the interaction of recombinant human SEMA3A and recombinant human NRP1. Briefly, SEMA3A were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to NRP1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-SEMA3A pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Activity Data (The interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. Upon activation by IL4, Th2 cells subsequently produce additional IL4 in a positive feedback loop.
