The IFNa proteins are produced by leukocytes. They are mainly involved in innate immune response against viral infection. It is also made synthetically as medication in hairy cell leukemia. Besides, Interferon Alpha/Beta Receptor 1 (IFNa/bR1) has been identified as an interactor of IFNa, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat IFNa and recombinant rat IFNa/bR1. Briefly, IFNa were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IFNa/bR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IFNa pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. Activity Data (High Mobility Group Protein 1 (HMG1) is among the most important chromatin proteins. In the nucleus HMGB1 interacts with nucleosomes, transcription factors, and histones. This nuclear protein organizes the DNA and regulates transcription. Activity Data (FGF13 (Fibroblast growth factor 13) is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth, and invasion. A proliferation assay was conducted to detect the bioactivity of recombinant rat FGF13 using 3T3 cells. Briefly, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of FGF13. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of 3T3 cells after incubation with FGF13 for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant FGF13 for 48h. The result was shown in Figure 2. It was obvious that FGF13 significantly increased cell viability of 3T3 cells.)

Activity Data (Myostatin (MSTN) also known as growth differentiation factor 8(GDF-8) is a member of the TGF beta protein family. Myostatin is aa secreted growth differentiation factor that produced and released by myocytes. This protein negatively regulates skeletal muscle cell proliferation and differentiation. Besides, Bone Morphogenetic Protein 1 (BMP1) has been identified as an interactor of MSTN, thus a binding ELISA assay was conducted to detect the interaction of recombinant human MSTN and recombinant human BMP1. Briefly, MSTN were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to BMP1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-MSTN pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of MSTN and BMP1 was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Interferon gamma (IFNgamma) is a dimerized soluble cytokine that is the only member of the type II class of interferons. The importance of IFNgamma in the immune system stems in part from its ability to inhibit viral replication directly, and most importantly from its immunostimulatory and immunomodulatory effects. It has been reported that IFN-gamma promotes production of inducible Nitric Oxide Synthase (iNOS) in macrophages as an important activator. After stimulated with IFN-gamma, morphological changes will occur in murine macrophage cell line (Raw 264.7 cells), and inducible nitric-oxide synthase (iNOS) in the cells will increase. Activity Data (FGF22 (Fibroblast growth factor 22) is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities and are involved in a variety of biological processes including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. A proliferation assay was conducted to detect the bioactivity of recombinant mouse FGF22 using 3T3 cells. Briefly, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of FGF22. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of 3T3 cells after incubation with FGF22 for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant FGF22 for 48h. The result was shown in Figure 2. It was obvious that FGF22 significantly increased cell viability of 3T3 cells.)

Activity Data (Arginase (Arg) is an enzyme that catalyzes the degradation of arginine to produce urea and omithine, which is crucial in the urea cycle.
