Activity Data (FAS (Tumor necrosis factor receptor superfamily member 6) belongs to the tumor necrosis factor receptor superfamily. FAS contains a death domain, which has been shown to play a central role in the physiological regulation of programmed cell death. A binding ELISA assay was conducted to detect the association of FAS with TNFa. Briefly, FAS were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL FAS were then transferred to TNFa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-FAS pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of FAS and TNFa was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Figure. The binding activity of TAT with GS.Tyrosine aminotransferase (TAT) is an enzyme present in the liver and catalyzes the conversion of tyrosine to 4-hydroxyphenylpyruvate. In humans, the tyrosine aminotransferase protein is encoded by the TAT gene. A deficiency of the enzyme in humans can result in what is known as Type II Tyrosinemia, wherein there is an abundance of tyrosine as a result of tyrosine failing to undergo an aminotransferase reaction to form 4-hydroxyphenylpyruvate. Besides, Glutamine synthetase (GS) has been identified as an interactor of TAT, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse TAT and recombinant mouse GS. Briefly, TAT were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to GS-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Figure. The binding activity of Slit2 with GREM1.Slit is a family of secreted extracellular matrix proteins which play an important signalling role in the neural development of most bilaterians. Humans, mice and other vertebrates possess three Slit homologs: Slit1, Slit2 and Slit3. The Slit2 protein has recently been discovered to be associated with the development of new blood vessels from pre-existing vessels, or angiogenesis. Slit2 has been implicated in promoting angiogenesis in mice (both in vitro and in vivo), in the human placenta, and in tumorigenesis. Besides, Gremlin 1 (GREM1) has been identified as an interactor of Slit2, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat Slit2 and recombinant rat GREM1. Briefly, Slit2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to GREM1-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Leptin is a hormone made by adipose cells that helps to regulate energy balance by inhibiting hunger. Many of leptin's effects are mediated through neuropeptide- containing neurons and neuropeptide receptors in the hypothalamus. Although regulation of fat stores is deemed to be the primary function of leptin, it also plays a role in other physiological processes, as evidenced by its multiple sites of synthesis other than fat cells, and the multiple cell types beside hypothalamic cells that have leptin receptors. Besides, Hexokinase 3, White Cell (HK3) has been identified as an interactor of LEP, thus a binding ELISA assay was conducted to detect the interaction of recombinant human LEP and recombinant human HK3. Briefly, LEP were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HK3-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Interleukin-6 (IL-6), a pro-inflammatory cytokine and an anti-inflammatory myokine, plays important roles in the acute phase reaction, inflammation, hematopoiesis, bone metabolism, and cancer progression. It has been reported that IL-6 significantly increased the MMP-10 protein lever in the human lung cancer A549 cells through the JAK/STAT signaling pathway. Briefly, A549 cells were seeded into 6-well cell culture clusters and allowed to grow to 50-70% confluence, then different concentrations of IL-6 was added. After incubated for 24h, the protein levels of MMP-10 in the cell supernatant were determined by Western blot.MMP-10 protein lever significantly increased in A549 cells due to the stimulation of IL-6, the data was shown in Figure 1.)

Activity Data (Matrix Metalloproteinase 13 (MMP13) is a member of the matrix metalloproteinase (MMP) family. MMP13 has been proposed to participate in aggrecan degradation associated with osteoarthritis and cleavage of type II collagen in osteoarthritic cartilage explants and in tumor progression and metastasis. MMP13 is likely to play a crucial role in the modulation of extracellular matrix degradation and cell-matrix interactions. In addition, it can cleave type I, III, IV, IX, X and XIV collagens and fibronectin. Thus we have chosed casein-zymography to measure the activity of MMP13. Briefly, various concentrations of MMP13 (10ug, 5ug, 1ug, 0.1ug, 0.01ug) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphat-polyacrylamide gel (SDS-PAGE; 15% gels) containing casein (1 mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP13 would hydrolyze casein nearby, which was indicated by the white bands on the gel. In this experiment we use heat-denatured MMP13 protein as negative control, and trypsase (1ug/mL) as positive control.Result : Casein hydrolysis by recombinant human MMP13 was shown in figure 1.)

Activity Data (IL17D (Interleukin-17D) belongs to the IL17 (interleukin 17) family, which includes six members (IL-17, IL-17B through IL-17F). IL17D is reported to induce the expression of IL6, CXCL8/IL8, and CSF2/GM-CSF from endothelial cells. IL17 family members share some similarities in AA sequence and structure, thus, a binding ELISA assay was constructed to detect the association of recombinant human IL17D with recombinant human IL17RB. Briefly, IL17D were diluted serially in PBS with 0.1%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17RB-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17D pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL17D with IL17RB was shown in Figure 1 and this effect was in a dose dependent manner.)

Activity Data (Figure. The binding activity of IL18 with CASP1.Interleukin-18 (IL18, also known as interferon-gamma inducing factor) is a proinflammatory cytokine that belongs to the IL-1 superfamily and is produced by macrophages and other cells. IL18 works by binding to the interleukin-18 receptor, and together with IL-12 it induces cell-mediated immunity following infection with microbial products like lipopolysaccharide (LPS). Besides, Caspase 1 (CASP1) has been identified as an interactor of IL18, thus a binding ELISA assay was conducted to detect the interaction of recombinant human IL18 and recombinant human CASP1. Briefly, IL18 were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CASP1-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Signal transducer and activator of transcription 3 (STAT3) is a member of the STAT protein family. STAT3 mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis. STAT3 is phosphorylated by receptor-associated Janus kinases (JAK), form homo- or heterodimers, and translocate to the cell nucleus where they act as transcription activators. Besides, Epidermal Growth Factor Receptor 2 (EGFR2) has been identified as an interactor of STAT3, thus a binding ELISA assay was conducted to detect the interaction of recombinant human STAT3 and recombinant human EGFR2. Briefly, STAT3 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to EGFR2-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-STAT3 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of STAT3 and EGFR2 was shown in Figure 1, and this effect was in a dose dependent manner.Figure. Activity Data (IL1R1 (Interleukin 1 Receptor Type I), also known as CD121a, is an important mediator involved in many cytokine induced immune and inflammatory responses. It belongs to the interleukin-1 receptor family, and is a receptor for interleukin 1 alpha (IL1A), interleukin 1 beta (IL1B), and interleukin 1 receptor antagonist (IL1RA). Besides, mouse IL1B shares 87.0% AA sequence identity with rat IL1B, suggesting the exist of cross-species activity. Thus, a binding ELISA assay was constructed to detect the association of recombinant rat IL1R1 with recombinant mouse IL1B. Briefly, IL1R1 were diluted serially in PBS with 0.1%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL1B-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL1R1 pAb, then aspirated and washed 3 times. Activity Data (HIF1a (Hypoxia-inducible factor 1-alpha) is a subunit of a transcription factor, which functions as a master transcriptional regulator of the adaptive response to hypoxia. The protein chaperone heat shock protein 90 (Hsp90) is a major regulator of different transcription factors, including HIF1a, thus a binding ELISA assay was conducted to detect the interaction of HIF1a and HSP90. Briefly, recombinant human HIF1a were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HSP90-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (PDGFA (Platelet-derived growth factor subunit A) is a Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. PDGFA has been described as a chemoattractant for monocytes and proven to be able to induce chemotactic migration of THP-1 cells. Therefore, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of PDGFA on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension,106cells/mL in RPMI 1640 with 0.5% FBS) and PDGFA (1.5ng/mL, 7.5ng/mL and 15ng/mL diluted separately in serum free RPMI 1640 ) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37oC with 5%CO2 for 3h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×40) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Result shows PDGFA is able to induce migration of THP-1 cells. The migrated THP-1 cells in low chamber at low magnification (×40) were shown in Figure 1. Five fields of each chamber were randomly chosen, and the migrated cells were counted at high magnification (×400). Statistical results were shown in Figure 2. The optimum chemotaxis of PDGFA occurs at 1.5ng/mL.)
