SDS-Page

(Pull down assay: a) pull down of hIL-33-His by hST2-Fc; b) pull down of hST2-Fc by hIL-33-His: 5ug of hST2-Fc (or control Fc protein), 2ug of hIL-33-His (or control His protein), and protein G resin (or anti-His resin) were incubated in 0.5ml RIPA buffer overnight at 4°C. The precipitates were separated by SDS-PAGE, electro-transferred onto NC membrane, and immunoblotted for the presence of hIL-33-His or hST2-Fc with anti-His HRP or anti-hIgG HRP, respectively.)

Related Product Information for IL33 active protein
Interleukin-33 (IL-33; HF-NEV; IL-1F11), a member of the IL-1 family of cytokines, is expressed by many cell types following pro-inflammatory stimulation and is thought to be released on cell lysis. The 30kD human IL33 is converted by CASP1 to a 18kD protein. IL33 binds to and signals through ST2 (IL1R1) and its stimulation recruits MYD88, IRAK, IRAK4, and TRAF6, followed by phosphorylation of ERK1 (MAPK3)/ERK2 (MAPK1), p38 (MAPK14), and JNK. The ability of IL-33 to target numerous immune cell types, like Th2-like cells, mast cells, and B1 cells, and to induce cytokine and chemokine production underlines its potential in influencing the outcome of a wide range of diseases, such as arthritis, asthma, atopic allergy & anaphylaxis, cardiovascular disease/atherosclerosis, nervous system diseases, and sepsis.

NCBI Description

IL33 (MIM 608678) is a member of the IL1 (see MIM 147760) family that potently drives production of T helper-2 (Th2)-associated cytokines (e.g., IL4; MIM 147780). IL33 is a ligand for IL33R (IL1RL1; MIM 601203), an IL1 family receptor that is selectively expressed on Th2 cells and mast cells (summary by Yagami et al., 2010 [PubMed 20926795]).[supplied by OMIM, Jan 2011]

Uniprot Description

IL33: Cytokine that binds to and signals through IL1RL1/ST2 and its stimulation recruits MYD88, IRAK1, IRAK4, and TRAF6, followed by phosphorylation of MAPK3/ERK1 and/or MAPK1/ERK2, MAPK14, and MAPK8. Induces T-helper type 2-associated cytokines. Belongs to the IL-1 family. Highly divergent. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cytokine

Chromosomal Location of Human Ortholog: 9p24.1

Product Notes

The IL33 il33 (Catalog #AAA637686) is an Active Protein produced from Human, E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: Human IL-33 (aa 112-270) is fused at the C-terminus to a His-tag. It is sometimes possible for the material contained within the vial of "IL-33, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
