Biological Activity
Specific activity is > 5,000pmol/min/ug, and was obtained by measuring the decrease of NAD in absorbance at 340nm resulting from NADH at pH 6.5 at 37C.
Assay
1. Prepare 180ul assay buffer into a suitable container: The concentrations are 80 mM Triethanolamine (pH 6.5), 0.3 mM 2-phosphorglycerate, 0.2 mM beta- NADH, 25 mM magnesium sulfate, 100 mM potassium chloride, 1.2 mM ADP, 3.5 unit pyruvate kinase, 5 unit L-lactic dehydrogenas.
2. Equilibrate to 37C and monitor at A340nm until the value is constant using a spectrophotometer.
3. Add 20ul of recombinant ENO3 protein 50ug/ml in assay buffer.
4. Record the decrease in A340 nm for 20 minutes at 37C.
Preparation and Storage
Can be stored at 4 degree C short term (1-2 weeks).
For long term storage, aliquot and store at -20 degree C or -70 degree C.
Avoid repeated freezing and thawing cycles.

Related Product Information for ENO3 active protein
Beta-enolase, also known as ENO3, is one of the three enolase isoenzymes found in mammals. This isoenzyme, a homodimer, is found in skeletal muscle cells in the adult. ENO3 play a role in converting phosphoglyceric acid to phosphenolpyruvic acid in the glycolytic pathway. Mutations in its gene can be associated with metabolic myopathies that may result from decreased stability of the enzyme. Recombinant human ENO3 protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.

NCBI Description

This gene encodes one of the three enolase isoenzymes found in mammals. This isoenzyme is found in skeletal muscle cells in the adult where it may play a role in muscle development and regeneration. A switch from alpha enolase to beta enolase occurs in muscle tissue during development in rodents. Mutations in this gene have be associated glycogen storage disease. Alternatively spliced transcript variants encoding different isoforms have been described.[provided by RefSeq, Jul 2010]

Uniprot Description

Appears to have a function in striated muscle development and regeneration.

Product Notes

The ENO3 eno3 (Catalog #AAA204286) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is His-tag. AAA Biotech's ENO3 can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Researchers should empirically determine the suitability of the ENO3 eno3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: MGSSHHHHHH SSGLVPRGSH MAMQKIFARE ILDSRGNPTV EVDLHTAKGR FRAAVPSGAS TGIYEALELR DGDKGRYLGK GVLKAVENIN STLGPALLQK KLSVADQEKV DKFMIELDGT ENKSKFGANA ILGVSLAVCK AGAAEKGVPL YRHIADLAGN PDLILPVPAF NVINGGSHAG NKLAMQEFMI LPVGASSFKE AMRIGAEVYH HLKGVIKAKY GKDATNVGDE GGFAPNILEN NEALELLKTA IQAAGYPDKV VIGMDVAASE FYRNGKYDLD FKSPDDPARH ITGEKLGELY KSFIKNYPVV SIEDPFDQDD WATWTSFLSG VNIQIVGDDL TVTNPKRIAQ AVEKKACNCL LLKVNQIGSV TESIQACKLA QSNGWGVMVS HRSGETEDTF IADLVVGLCT GQIKTGAPCR SERLAKYNQL MRIEEALGDK AIFAGRKFRN PKAK. It is sometimes possible for the material contained within the vial of "ENO3, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
