Activity Data (Fibronectin type III domain-containing protein 5, the precursor of irisin, is a protein that is encoded by the FNDC5 gene. It was reported that FNDC5 significantly decreased cell number, migration and viability through apoptosis in malignant MDA-MB-231 cells. Thus MDA-MB-231 cells were seeded overnight at a density of 5,000 cells/well, and treated with or without various concentrations of FNDC5 for 48h, then MDA-MB-231 cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8). The binding activity of FGF2 with CASP1.Basic fibroblast growth factor (FGF2), also known as bFGF, FGF-beta is a member of a large family of structurally related heparin-binding proteins (the FGFs) involved in the regulation of cell proliferation, growth and differentiation. It involved in many biological processes including angiogenesis, embryonic development and wound healing. Additionally, FGF2 is a critical component of human embryonic stem cell culture medium. Besides, Caspase 1 (CASP1) has been identified as an interactor of FGF2, thus a binding ELISA assay was conducted to detect the interaction of recombinant bovine FGF2 and recombinant bovine CASP1. Briefly, FGF2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CASP1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-FGF2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. Activity Data (IL24 (interleukin 24) is a cytokine that belongs to IL10 family. This protein can induce apoptosis selectively in various cancer cells, including ECV304. Thus, inhibition of cell proliferation assay of IL24 was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640 prior to the addition of various concentrations of IL24. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Activity Data (IL18 (interleukin 18) is a cytokine that shares similarities in structure with IL1, and is produced by macrophages and other cells. IL18 has been reported to stimulate T cells proliferation and NK cell activity, besides, murine IL18 has been found to enchance the production of INFgamma in spleen cells in vitro. Activity Data (Lactoferrin (LF), also known as lactotransferrin (LTF), is a multifunctional protein of the transferrin family. Activity Data (Mechanism: MMP9 is a zinc-dependent enzymes capable of cleaving components of the extracellular matrix, which belongs to the matrix metalloproteinase (MMP) family. It is a gelatinase A, 92kDa type IV collagenase which can hydrolyze gelatin under certain conditions. Activity Data (TF-1, the human erythroleukemia cell line,provides a good system for detecting the activity of GM-CSF for it is a cell line of immature erythroid origin that completely depends on interleukin 3 (IL-3) or granulocyte-macrophage colony-stimulating factor (GM-CSF) for long term growth. As reported, GM-CSF was also able to induce differentiation of human monoblastic leukemia cell line U937.
