(IL6R (Interleukin-6 receptor subunit alpha) is part of the receptor for interleukin 6. The interaction of IL6R and IL6 may lead to the regulation of the immune response, acute-phase reactions and hematopoiesis. Thus, a binding ELISA assay was conducted to detect the association of IL6R with IL6. Briefly, IL6R were diluted serially in PBS with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL6-coated microtiter wells and incubated for 2h at 37 degree C. (Insulin-like growth factor I (IGF1), is a hormone similar in molecular structure to insulin but have a much higher growth-promoting activity, it belongs to a family of proteins involved in mediating growth and development. It is reported that IGF1 induces the proliferation, migration, differentiation of a large types of cells including the MCF-7 breast cancer cell line. To test the effect of growth factors on proliferation, MCF-7 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of IGF-1. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). (Vascular endothelial growth factor C (VEGFC) is a protein that is a member of the platelet-derived growth factor/vascular endothelial growth factor (PDGF/VEGF) family. It plays key roles in the physiology and pathology of many aspects of the cardiovascular system, including vasculogenesis, hematopoiesis, angiogenesis and vascular permeability. (NEU (Sialidase-1) is an enzyme that catalyzes the removal of sialic acid (N-acetylneuraminic acid) moities from glycoproteins and glycolipids. In the lysosome, this enzyme is part of a heterotrimeric complex together with beta-galactosidase and cathepsin A (CTSA). Thus a binding ELISA assay was conducted to detect the interaction of recombinant human NEU and recombinant human CTSA. Briefly, NEU were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CTSA-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-NEU pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of NEU and CTSA was shown in Figure 1, and this effect was in a dose dependent manner.)
