Biological Activity
Specific activity is > 210 nmoles/min/ug, and is defined as the amount of enzyme that cleaves 1umole of suc-AAFP-pNA per minute at 1C in Tris-Hcl pH8.0 using chymotrypsin.
Assay
1. Prepare assay buffer into a suitable container and pre-chilling on ice before use: The final concentrations are 35 mM Tris, pH 8.0, 1uM suc-AAFP-pNA).
2. Add recombinant Cyclophilin E protein with various concentrations (1ug, 2ug, 5ug) in assay buffer.
3. Mix by inversion and equilibrate to 1C and monitor the A410nm until the value is constant using a spectrophotometer.
4. Add pre-chilled chymotrypsin to 12.5 uM and mix immediately.
5. Record the increase in A410 nm for 20 minutes
Preparation and Storage
Can be stored at 4 degree C short term (1-2 weeks).
For long term storage, aliquot and store at -20 degree C or -70 degree C.
Avoid repeated freezing and thawing cycles.

Related Product Information for PPIE active protein
Cyclophilins possess the activity of peptidyl-prolyl cis-trans isomerase (PPIase) and are involved in cellular protein folding and protein interactions. PPIE contains two RNA binding domains at the N-terminal region and a PPIase domain at the C-terminal region. It was not known what cellular and physiological roles of PPIE. Recombinant human PPIE was expressed in E Coli and purified by conventional chromatography techniques.

NCBI Description

The protein encoded by this gene is a member of the peptidyl-prolyl cis-trans isomerase (PPIase) family. PPIases catalyze the cis-trans isomerization of proline imidic peptide bonds in oligopeptides and accelerate the folding of proteins. This protein contains a highly conserved cyclophilin (CYP) domain as well as an RNA-binding domain. It was shown to possess PPIase and protein folding activities, and it also exhibits RNA-binding activity. Alternative splicing results in multiple transcript variants. A related pseudogene, which is also located on chromosome 1, has been identified. [provided by RefSeq, Aug 2010]

Uniprot Description

PPIases accelerate the folding of proteins. It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. Combines RNA-binding and PPIase activities. May be involved in muscle- and brain-specific processes. May be involved in pre-mRNA splicing.

Product Notes

The PPIE ppie (Catalog #AAA203257) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is His-tag. AAA Biotech's PPIE (Cyclophilin E) can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Researchers should empirically determine the suitability of the PPIE ppie for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: MRGSHHHHHH GMASMTGGQQ MGRDLYDDDD KDRWGSMATT KRVLYVGGLA EEVDDKVLHA AFIPFGDITD IQIPLDYETE KHRGFAFVEF ELAEDAAAAI DNMNESELFG RTIRVNLAKP MRIKEGSSRP VWSDDDWLKK FSGKTLEENK EEEGSEPPKA ETQEGEPIAK KARSNPQVYM DIKIGNKPAG RIQMLLRSDV VPMTAENFRC LCTHEKGFGF KGSSFHRIIP QFMCQGGDFT NHNGTGGKSI YGKKFDDENF ILKHTGPGLL SMANSGPNTN GSQFFLTCDK TDWLDGKHVV FGEVTEGLDV LRQIEAQGSK DGKPKQKVII ADCGEYV. It is sometimes possible for the material contained within the vial of "PPIE (Cyclophilin E), Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
