(Platelet Derived Growth Factor Subunit A (PDGFA) is a member of Platelet-derived growth factor (PDGF) family which plays a significant role in blood vessel formation, the growth of blood vessels from already-existing blood vessel tissue, mitogenesis, i.e. proliferation, of mesenchymal cells such as fibroblasts, osteoblasts, tenocytes, vascular smooth muscle cells and mesenchymal stem cells as well as chemotaxis, the directed migration, of mesenchymal cells. Besides, Platelet Derived Growth Factor Receptor Alpha (PDGFRa) has been identified as an interactor of PDGFA, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PDGFA and recombinant human PDGFRa. Briefly, PDGFA were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100mul were then transferred to PDGFRa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PDGFA pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of PDGFA and PDGFRa was shown in Figure 1, and this effect was in a dose dependent manner.Figure. (Figure. The binding activity of CTSD with HSP90b1.Cathepsin D (CSTD) is an aspartic protease that depends critically on protonation of its active site Asp residue. Along with Asp-protonation, lower pH also leads to conformational switch in cathepsin-D: the N-terminal segment of the protease moves out of the active site as pH drops. Similar to other aspartic protainases, cathepsin D accommodates up to 8 amino acid residues in the binding cleft of the active site. (Nerve growth factor (NGF) is a neurotrophic factor and neuropeptide primarily involved in the regulation of growth, maintenance, proliferation, and survival of certain target neurons. NGF is also involved in pathways besides those regulating the life cycle of neurons. a nerve growth factor (NGF)-dependent TF-1 subclone cell line suitable for determination of bioactivity of recombinant NGF(rhNGF), then TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 contain various concentrations of recombinant rat NGF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of TF-1 cells after incubation with TF-1 for 72h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 assay after incubation with recombinant NGF for 72h. The result was shown in Figure 2. (Regenerating Islet Derived Protein 3 Beta (REG3b) also known as PAP-I and HIP is bactericidal C-type lectin which acts against several intestinal Gram-positive bacteria and Gram-negative bacteria. The Reg family proteins have been implicated in a range of physiological processes including acting as acute phase reactants, lectins, survival/growth factors for insulin-producing pancreatic beta-cells, neural cells, and epithelial cells of the digestive system. To test the effect of REG3b on cell proliferation of SK-N-SH, SK-N-SH cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of REG3b. (IL24 (interleukin 24) is a cytokine that belongs to IL10 family. This protein can induce apoptosis selectively in various cancer cells, including ECV304. Thus, inhibition of cell proliferation assay of IL24 was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640 prior to the addition of various concentrations of IL24. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). (Epidermal growth factor (EGF) is a growth factor that stimulates cell growth, proliferation, and differentiation by binding to its receptor EGFR. To test the effect of EGF on cell proliferation of 3T3 fibroblasts, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2, 000cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of EGF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37 degree C .Cell proliferation of 3T3 cells after incubation with EGF for 72h observed by inverted microscope was shown in Figure 1.)

(IL18 (interleukin 18) is a cytokine that shares similarities in structure with IL1, and is produced by macrophages and other cells. IL18 has been reported to stimulate T cells proliferation and NK cell activity, besides, murine IL18 has been found to enchance the production of INFgamma in spleen cells in vitro. Thus, a stimulation assay was conducted to detect the activity of IL18 using murine spleen cells. Briefly, murine spleen cells were seeded into wells of 6-well plates at a density of 2×106 cells/mL in RPMI-1640 with the addition of various concentrations of IL18. After incubation for 48 hours, the concentration of INFgamma in the cell supernatant was detected using an ELISA kit. INFgamma levels in the cell supernatant of spleen cells increased significantly after stimulated with IL18, the data was shown in Figure 1.)
