Activity Data (NOG (Noggin) is a signaling molecule that plays an important role in promoting somite patterning in the developing embryo, essential for cartilage morphogenesis and joint formation. It is considered as an inhibitor in bone morphogenetic proteins (BMP) signaling, by binding with BMP4, thus a binding ELISA assay was conducted to detect the association of recombinant rat NOG with BMP4. Briefly, NOG were diluted serially in PBS with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to BMP4-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-NOG pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of NOG with BMP4 was shown in Figure 1 and this effect was in a dose dependent manner.)

The ornithine decarboxylation reaction catalyzed by ornithine decarboxylase is the first and committed step in the synthesis of polyamines, particularly putrescine, spermidine and spermine. Polyamines are important for stabilizing DNA structure, the DNA double strand-break repair pathway and as antioxidants. Therefore, ornithine decarboxylase is an essential enzyme for cell growth, producing the polyamines necessary to stabilize newly synthesized DNA. Lack of ODC causes cell apoptosis in embryonic mice, induced by DNA damage. Besides, Thymidine Kinase 1, Soluble (TK1) has been identified as an interactor of ODC, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ODC and recombinant human TK1. Activity Data (Figure. Cell proliferation of Raw264.7 cells after stimulated with SCF.Stem cell factor (also known as SCF, KIT-ligand, KL, or steel factor) is a cytokine that binds to the c-KIT receptor (CD117). SCF can exist both as a transmembrane protein and a soluble protein. This cytokine plays an important role in hematopoiesis (formation of blood cells), spermatogenesis, and melanogenesis. SCF has been shown to  stimulate the proliferation of Raw264.7 cells. To test this effect, Raw264.7 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach later, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM containing various concentrations of recombinant mouse SCF. After incubated for 120h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of Raw264.7 cells after incubation with SCF for 120h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant SCF for 120h. The result was shown in Figure 2. Activity Data (Interleukin-2 (IL-2), the main lymphokine produced by activated CD4 T-helper lymphocytes, is a potent T-cell growth regulator. In addition to this activity, IL-2 was found to decrease the proliferation of 3T3 fibroblasts in time- and dose-dependent manner. Activity Data (Figure. The binding activity of VEGF165 with VEGFR1.Vascular endothelial growth factor 165 is one kind of isoforms of Vascular endothelial growth factor A (VEGFA). This protein is a glycosylated mitogen that specifically acts on endothelial cells and has various effects, including mediating increased vascular permeability, inducing angiogenesis, vasculogenesis and endothelial cell growth, promoting cell migration, and inhibiting apoptosis. Besides, Vascular Endothelial Growth Factor Receptor 1 (VEGFR1) has been identified as an interactor of VEGF165, thus a binding ELISA assay was conducted to detect the interaction of recombinant human VEGF165 and recombinant human VEGFR1. Briefly, VEGF165 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to VEGFR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-VEGF165 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Activity Data (SDF1 (stromal cell-derived factor 1), also known as C-X-C motif chemokine 12, is a chemokine protein that has chemotaxis active on T-lymphocytes and monocytes. It is thought that SDF1 stimulates migration of monocytes and T-lymphocytes through its receptors, CXCR4 and ACKR3; thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of SDF1 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension,106cells/mL in RPMI 1640 with 0.5% FBS) and SDF1 (10ng/mL, 30ng/mL and 60ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37 degree C with 5% CO2 for 3h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×100) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Result shows SDF1 is able to induce migration of THP-1 cells. The migrated THP-1 cells in low chamber at low magnification (×100) were shown in Figure 1. Five fields of each chamber were randomly chosen, and the migrated cells were counted at high magnification (×400). Statistical results were shown in Figure 2. The optimum chemotaxis of SDF1 occurs at 10ng/mL.)
