Activity Data (Figure. The binding activity of TK1 with GAPDH.Thymidine kinase 1 (TK1), is a human thymidine kinase. Thymidine kinase has been making a growing impact in the cancer research community. It has been found that elevated blood serum levels of TK-1 correlates with metastatic capabilities of the cancer and thereby can be used to detect malignant types of cancer, furthermore TK-1 has been found to show up in blood serum even before clinical symptoms even start to show. Besides, Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) has been identified as an interactor of TK1, thus a binding ELISA assay was conducted to detect the interaction of recombinant human TK1 and recombinant human GAPDH. Briefly, TK1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to GAPDH-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TK1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TK1 and GAPDH was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Lactoferrin (LF), also known as lactotransferrin (LTF), is a multifunctional protein of the transferrin family. Lactoferrin is one of the components of the immune system of the body; it has antimicrobial activity (bacteriocide, fungicide) and is part of the innate defense, mainly at mucoses. In particular, lactoferrin provides antibacterial activity to human infants. Lactoferrin interacts with DNA and RNA, polysaccharides and heparin, and shows some of its biological functions in complexes with these ligands. LTF is known to activate immune cells to produce several cytokines, such as tumor necrosis factor TNF-a, thus, a stimulation assay was conducted to detect the activity of LTF using human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into wells of 24-well plates at a density of 5×106 cells/mL in RPMI-1640 with the addition of various concentrations of recombinant human LTF. After incubation for 20 hours, the concentration of TNF-a in the cell supernatant was detected using an ELISA kit. Testing Data (Cathepsin V (CTSV) is a lysosomal cysteine proteinase which belongs to peptidase C1 family. It is expressed in normal human thymus, testis and corneal epithelium. Cathepsin V plays an important role in corneal physiology and mediates degradation of invariant chain in human thymus. Besides, Retinoblastoma Protein 1 (RB1) has been identified as an interactor of CTSV, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CTSV and recombinant human RB1. Briefly, CTSV were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to RB1-coated microtiter wells and incubated for 2h at 37°C. Wells were washed with PBST and incubated for 1h with anti-CTSV pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37°C. Finally, add 50µL stop solution to the wells and read at 450nm immediately. The binding activity of CTSV and RB1 was shown in Figure 1, and this effect was in a dose dependent manner.Figure 1.The binding activity of CTSV with RB1.)

Activity Data (Mechanism: MMP9 is a zinc-dependent enzymes capable of cleaving components of the extracellular matrix, which belongs to the matrix metalloproteinase (MMP) family. It is a gelatinase A, 92kDa type IV collagenase which can hydrolyze gelatin under certain conditions. Gelatin zymography is mainly used for the detection of the gelatinases, MMP-2 and MMP-9 and It is extremely sensitive because levels of 10pg of MMP-2 can already be detected. Briefly, various concentrations of recombinant human MMP9 (1000ng, 500ng, 100ng, 10ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphate- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP2 would hydrolyze gelatin nearby, which was indicated by the white binds on the gel. In this experiment we use heat-denatured MMP9 protein as negative control, and blood sample as positive control. Result: Gelatin hydrolysis by recombinant human MMP9 (10-70kd) was shown in figure 1.)
