(IL11 (Interleukin-11) is a multifunctional cytokine first isolated from bone marrow-derived stromal cells. It stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells and induces megakaryocyte maturation resulting in increased platelet production. Besides, IL11 is reported to induce the proliferation of human T-cells, thus, a proliferation assay was conducted to detect the bioactivity of human recombinant IL11 using Jurkat cells. Briefly, Jurkat cells were seeded into triplicate wells of 96-well plates at a density of 10, 000 cells/well in RPMI-1640 with the addition of various concentrations of IL11. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Cell proliferation of Jurkat cells after incubation with IL11 for 72h observed by inverted microscope was shown in Figure 1. The CCK-8 data was shown in Figure 2. It was obvious that IL11 significantly promoted cell proliferation of Jurkat cells.)

The binding activity of MIF with MHCDG.Macrophage migration inhibitory factor (MIF), also known as glycosylation-inhibiting factor (GIF), L-dopachrome isomerase, or phenylpyruvate tautomerase is a protein classified as an inflammatory cytokine. MIF is an important regulator of innate immunity. It involved in cell-mediated immunity, immunoregulation, and inflammation. MIF plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. This lymphokine and the JAB1 protein form a complex in the cytosol near the peripheral plasma membrane, which may indicate a role in integrin signaling pathways. Besides, Major Histocompatibility Complex Class II Invariant Chain (MHCDG) has been identified as an interactor of MIF, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse MIF and recombinant mouse MHCDG. Briefly, MIF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to MHCDG-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-MIF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of MIF and MHCDG was shown in Figure 1, and this effect was in a dose dependent manner.)

(Macrophage Colony Stimulating Factor (M-CSF), also known as CSF-1,is a secreted cytokine which influences hematopoietic stem cells to differentiate into macrophages or other related cell types. M-CSF (or CSF-1) is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. It can also affects macrophages and monocytes in several ways, including stimulating increased phagocytic and chemotactic activity, and increased tumour cell cytotoxicity. The role of M-CSF is not only restricted to the monocyte/macrophage cell lineage. By interacting with its membrane receptor (CSF1R or M-CSF-R encoded by the c-fms proto-oncogene), M-CSF also modulates the proliferation of earlier hematopoietic progenitors and influence numerous physiological processes involved in immunology, metabolism, fertility and pregnancy. (Vascular endothelial growth factor A (VEGF-A), a glycosylated mitogen, is known to be a vascular permeability factor and an endothelial cell growth factor secreted by the smooth muscle and endothelial cells. It has been reported that VEGF-A induces vascular permeability and growth, promotes monocyte/macrophage migration, and inhibits cell apoptosis and so on. To test the effect of VEGF-A on cell proliferation of ECV304 endothelium cell line, cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of VEGFA. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC .Cell proliferation of ECV304 cells after incubation with VEGFA for 72h observed by inverted microscope was shown in Figure 1.)

(Figure. The binding activity of IGF1 with IGFBP3.Insulin-like growth factor 1 (IGF1), also called somatomedin C is a hormone similar in molecular structure to insulin. It plays an important role in childhood growth and continues to have anabolic effects in adults. A synthetic analog of IGF1, mecasermin, is used for the treatment of growth failure. Besides, Insulin Like Growth Factor Binding Protein 3 (IGFBP3) has been identified as an interactor of IGF1, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse IGF1 and recombinant mouse IGFBP3. Briefly, IGF1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to IGFBP3-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IGF1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. (Protein S100B is a member of the S100 family. S100 proteins are EF-hand calcium-binding proteins and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. Experimental results suggest that the receptor for advanced glycation end products (RAGE) plays important roles in mediating S100 protein-induced cellular signaling. Besides, rat RAGE shares similarities with human RAGE in amino acids sequence with the identity of 80.0%.
