Biological Activity
Specific activity is > 90,000 pmol/min/ug, and is defined as the amount of enzyme that conjugate 1.0 u mole of 1-chloro-2,4-dinitrobenzene (CDNB) with reduced glutathione per minute at pH 6.5 at 25C.
Assay
1. Prepare a 180ul assay buffer into a suitable container. The concentrations are 106mM potassium phosphate (pH6.5), 1.1mM EDTA, 5.5mM glutathione, reduced, 2.2mM 1-chloro-2,4-dinitrobenzene (CDNB), 8% (v/v) ethanol.
2. Equilibrate to 25C and monitor at A340nm until the value is constant using a spectrophotometer.
3. Add 20ul of recombinant GSTM5 protein 1.25ug/ml in assay buffer.
4. Mix by inversion and record the decrease at A340nm for approximately 5 minutes.
Preparation and Storage
Can be stored at 4 degree C short term (1-2 weeks).
For long term storage, aliquot and store at -20 degree C or -70 degree C.
Avoid repeated freezing and thawing cycles.

Glutathione S-transferase mu 5, also known as GSTM5, is member of the glutathione s-transferase (GST) family of proteins. There are eight families of GST proteins, namely alpha, kappa, mu, omega, pi, sigma, theta and zeta, each of which is composed of proteins that have a variety of functions throughout the cell. GSTM5 plays an important role in detoxification. Recombinant human GSTM5 protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.

NCBI Description

Cytosolic and membrane-bound forms of glutathione S-transferase are encoded by two distinct supergene families. At present, eight distinct classes of the soluble cytoplasmic mammalian glutathione S-transferases have been identified: alpha, kappa, mu, omega, pi, sigma, theta and zeta. This gene encodes a glutathione S-transferase that belongs to the mu class. The mu class of enzymes functions in the detoxification of electrophilic compounds, including carcinogens, therapeutic drugs, environmental toxins and products of oxidative stress, by conjugation with glutathione. The genes encoding the mu class of enzymes are organized in a gene cluster on chromosome 1p13.3 and are known to be highly polymorphic. These genetic variations can change an individual's susceptibility to carcinogens and toxins as well as affect the toxicity and efficacy of certain drugs. Diversification of these genes has occurred in regions encoding substrate-binding domains, as well as in tissue expression patterns, to accommodate an increasing number of foreign compounds.
