(Figure. Cell proliferation of Raw264.7 cells after stimulated with SCF.Stem cell factor (also known as SCF, KIT-ligand, KL, or steel factor) is a cytokine that binds to the c-KIT receptor (CD117). SCF can exist both as a transmembrane protein and a soluble protein. This cytokine plays an important role in hematopoiesis (formation of blood cells), spermatogenesis, and melanogenesis. SCF has been shown to stimulate the proliferation of Raw264.7 cells. To test this effect, Raw264.7 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach later, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM containing various concentrations of recombinant mouse SCF. After incubated for 120h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of Raw264.7 cells after incubation with SCF for 120h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant SCF for 120h. The result was shown in Figure 2. (Interleukin-2 (IL-2), the main lymphokine produced by activated CD4 T-helper lymphocytes, is a potent T-cell growth regulator. In addition to this activity, IL-2 was found to decrease the proliferation of 3T3 fibroblasts in time- and dose-dependent manner. Thus 3T3 cells were seeded overnight at a density of 5,000 cells/well, and treated with or without various concentrations of IL2 for 48h, then 3T3 cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8). (Figure. The binding activity of VEGF165 with VEGFR1.Vascular endothelial growth factor 165 is one kind of isoforms of Vascular endothelial growth factor A (VEGFA). This protein is a glycosylated mitogen that specifically acts on endothelial cells and has various effects, including mediating increased vascular permeability, inducing angiogenesis, vasculogenesis and endothelial cell growth, promoting cell migration, and inhibiting apoptosis. Besides, Vascular Endothelial Growth Factor Receptor 1 (VEGFR1) has been identified as an interactor of VEGF165, thus a binding ELISA assay was conducted to detect the interaction of recombinant human VEGF165 and recombinant human VEGFR1. Briefly, VEGF165 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to VEGFR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-VEGF165 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of VEGF165 and VEGFR1 was shown in Figure 1, and this effect was in a dose dependent manner.)

(SDF1 (stromal cell-derived factor 1), also known as C-X-C motif chemokine 12, is a chemokine protein that has chemotaxis active on T-lymphocytes and monocytes. It is thought that SDF1 stimulates migration of monocytes and T-lymphocytes through its receptors, CXCR4 and ACKR3; thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of SDF1 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension,106cells/mL in RPMI 1640 with 0.5% FBS) and SDF1 (10ng/mL, 30ng/mL and 60ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37 degree C with 5% CO2 for 3h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×100) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Result shows SDF1 is able to induce migration of THP-1 cells. The migrated THP-1 cells in low chamber at low magnification (×100) were shown in Figure 1. Five fields of each chamber were randomly chosen, and the migrated cells were counted at high magnification (×400). Statistical results were shown in Figure 2. The optimum chemotaxis of SDF1 occurs at 10ng/mL.)

The ED50 (median effective dose) for this effect is less than 0.25 ng/mL.In-house data of APA045Mu01 used in cellular experiment:Six-eight weeks old Balb/c mice were used for BMDC. At first, mouse femur and tibia were taken out, and then bone marrow was washed out with serum-free RPMI 1640 medium, followed by centrifugation at 1200RPM for 5min (4oC). ACK buffer was added to get rid of red blood cells, and then, centrifuged at 1200RPM for 5min (4oC). Cell pellets were collected and re-suspended, the cells were cultured in DMEM medium supplemented with 10% FBS, or DMEM medium supplemented with 10% FBS and GMCSF (APA045Mu01) at 37oC with 5% CO2 in thermostatic incubator. (Insulin (INS) is a polypeptide hormone originating in the beta cells of the pancreas and serving as a principal regulator for the storage and production of carbohydrates. Insulin decreases blood glucose concentration. It increases cell permeability to monosaccharides, amino acids and fatty acids. It accelerates glycolysis, the pentose phosphate cycle, and glycogen synthesis in liver. Besides, Insulin Receptor (ISR) has been identified as an interactor of INS, thus a binding ELISA assay was conducted to detect the interaction of recombinant bovine INS and recombinant human ISR. Briefly, INS were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to ISR-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-INS pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of INS and ISR was shown in Figure 1, and this effect was in a dose dependent manner.)
