Activity Data (Cytochrome P450 1A1 (CYP1A1) is a member of Cytochromes P450 superfamily of enzymes. Cytochromes P450 are a group of heme-thiolate monooxygenases. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. CYP1A1 is also known as AHH (aryl hydrocarbon hydroxylase). It is involved in the metabolic activation of aromatic hydrocarbons (polycyclic aromatic hydrocarbons, PAH). Besides, Heat Shock 70kDa Protein 4 (HSPA4) has been identified as an interactor of CYP1A1, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat CYP1A1 and recombinant rat HSPA4. Briefly, CYP1A1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HSPA4-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CYP1A1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of NAP3 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension, 106 cells/mL in RPMI 1640 with FBS free) and SLC (31.25ng/mL, 62.5ng/mL,125ng/mL and 250ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37 degree C with 5% CO2 for 1h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×100) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Result shows NAP3 is able to induce migration of THP-1 cells. The migrated THP-1 cells in low chamber at low magnification (×100) were shown in Figure 1. Activity Data (CCL13 (C-C motif chemokine 13) is a chemotactic factor that attracts monocytes, lymphocytes, basophils and eosinophils, which belongs to the CC chemokine subfamily. It has been reported that CCL13 can induce chemotactic migration of THP-1 cells. Activity Data (Figure. The binding activity of TGM1 with SNCa.Transglutaminase 1(TGM1) is an enzyme found in cells that make up the outermost layer of the skin (the epidermis). It can catalyze the addition of an alkyl group from an akylamine to a glutamine residue of a protein, forming an alkylglutamine in the protein. TGM1 is involved in the formation of the cornified cell envelope, which is a structure that surrounds skin cells and helps form a protective barrier between the body and its environment. Besides, Synuclein Alpha (SNCa) has been identified as an interactor of TGM1, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse TGM1 and recombinant mouse SNCa. Briefly, TGM1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to SNCa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TGM1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TGM1 and SNCa was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (INS (Insulin) is a peptide hormone produced by beta cells of the pancreatic islets, which decreases blood glucose concentration and increases cell permeability to monosaccharides, amino acids and fatty acids. It has been reported that insulin triggers phosphorylation of a number of substrates by binding to its receptors, which was important for cell proliferation, cell cycle progression, cell division and differentiation. Cell proliferation of TF-1 cells after stimulated with LIF.Leukemia Inhibitory Factor (LIF), is an interleukin 6 class cytokine that affects cell growth by inhibiting differentiation. Other properties attributed to the cytokine include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. To test the effect of LIF on cell proliferation, TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 including various concentrations of recombinant human LIF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of TF-1 cells after incubation with LIF for 72h observed by inverted microscope was shown in Figure 1.
