(Platelet Derived Growth Factor Subunit A (PDGFA) is a member of Platelet-derived growth factor (PDGF) family which plays a significant role in blood vessel formation, the growth of blood vessels from already-existing blood vessel tissue, mitogenesis, i.e. proliferation, of mesenchymal cells such as fibroblasts, osteoblasts, tenocytes, vascular smooth muscle cells and mesenchymal stem cells as well as chemotaxis, the directed migration, of mesenchymal cells. Besides, Platelet Derived Growth Factor Receptor Alpha (PDGFRa) has been identified as an interactor of PDGFA, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PDGFA and recombinant human PDGFRa. Briefly, PDGFA were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100mul were then transferred to PDGFRa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PDGFA pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of PDGFA and PDGFRa was shown in Figure 1, and this effect was in a dose dependent manner.Figure. (Nerve growth factor (NGF) is a neurotrophic factor and neuropeptide primarily involved in the regulation of growth, maintenance, proliferation, and survival of certain target neurons. NGF is also involved in pathways besides those regulating the life cycle of neurons. a nerve growth factor (NGF)-dependent TF-1 subclone cell line suitable for determination of bioactivity of recombinant NGF(rhNGF), then TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 contain various concentrations of recombinant rat NGF. (Regenerating Islet Derived Protein 3 Beta (REG3b) also known as PAP-I and HIP is bactericidal C-type lectin which acts against several intestinal Gram-positive bacteria and Gram-negative bacteria. The Reg family proteins have been implicated in a range of physiological processes including acting as acute phase reactants, lectins, survival/growth factors for insulin-producing pancreatic beta-cells, neural cells, and epithelial cells of the digestive system. To test the effect of REG3b on cell proliferation of SK-N-SH, SK-N-SH cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of REG3b. It was reported that FNDC5 significantly decreased cell number, migration and viability through apoptosis in malignant MDA-MB-231 cells. The binding activity of FGF2 with CASP1.Basic fibroblast growth factor (FGF2), also known as bFGF, FGF-beta is a member of a large family of structurally related heparin-binding proteins (the FGFs) involved in the regulation of cell proliferation, growth and differentiation. It involved in many biological processes including angiogenesis, embryonic development and wound healing. Additionally, FGF2 is a critical component of human embryonic stem cell culture medium. Besides, Caspase 1 (CASP1) has been identified as an interactor of FGF2, thus a binding ELISA assay was conducted to detect the interaction of recombinant bovine FGF2 and recombinant bovine CASP1. Briefly, FGF2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CASP1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-FGF2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of FGF2 and CASP1 was shown in Figure 1, and this effect was in a dose dependent manner.)

A) HeLa cells were treated with the ten compounds that produced the strongest reduction in pSer40/41MCM2 levels in the primary screen. Protein extracts were prepared and levels of residual pSer40/41MCM2 phosphorylation were measured by western blot analysis with the indicated antibodies. B) The same compounds were tested for their ability to inhibit CDC7 kinase activity in in vitro kinase assay.
