(Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. Current data suggest that direct application of IL-6 on breast cancer cells inhibits proliferation in ER-positive (estrogen- receptor- positive) cells through the Jak/Stat3 pathway. To test the inhibitory effect of IL-6 on proliferation of ER-positive MCF-7 cell line, cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of IL-6. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC.The inhibitory effect of IL-6 on cell proliferation of MCF-7 cells observed by inverted microscope and detected by CCK-8 was shown in Figure 1 and Figure 2 respectively (Dose-dependent effect was not detected in this case).)

(TNFa (Tumor necrosis factor), is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It has been reported that TNFa can inhibit the proliferation and induce apoptosis of A549 cells, besides, the concentration of IL-1beta and IL-8 in cell supernatant will increase after stimulation. Therefore, a stimulation assay of TNFa was conducted using A549 cells. Briefly, A549 cells were incubated in DMEM with different concentrations of TNFa (1ng/mL, 10ng/mL, 100ng/mL, 1000ng/mL) for 8h, after which the concentration of IL-1beta and IL-8 in the cell supernatant were detected by ELISA. IL-1beta and IL-8 levels in the cell supernatant of A549 cells increased significantly after stimulated with IL-1beta, the data was shown in Figure 1 and Figure 2 separately.)

(Fibronectin type III domain-containing protein 5, the precursor of irisin, is a protein that is encoded by the FNDC5 gene. It was reported that FNDC5 significantly decreased cell number, migration and viability through apoptosis in malignant MDA-MB-231 cells. Thus MDA-MB-231 cells were seeded overnight at a density of 5,000 cells/well, and treated with or without various concentrations of FNDC5 for 48h, then MDA-MB-231 cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8).
