Supplied as a lyophilized powder from 20mM Tris, 0.15M sodium chloride, pH 8. Reconstitute with sterile, ddH2O to a concentration of 0.1mg/ml.
Biological Activity
beta-Nerve Growth Factor is fully biologically active when compared to standards. The ED50, calculated by its ability to stimulate chick E9 DRG neurite outgrowth was found to be <1.0ng/ml, corresponding to a specific activity of >1x10e6 units/mg.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 1.1945 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of Recombinant beta-Nerve Growth Factor as a Reference Standard.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O, 0.1% HSA or BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

This gene is a member of the NGF-beta family and encodes a secreted protein which homodimerizes and is incorporated into a larger complex. This protein has nerve growth stimulating activity and the complex is involved in the regulation of growth and the differentiation of sympathetic and certain sensory neurons. Mutations in this gene have been associated with hereditary sensory and autonomic neuropathy, type 5 (HSAN5), and dysregulation of this gene's expression is associated with allergic rhinitis. Function: Nerve growth factor is important for the development and maintenance of the sympathetic and sensory nervous systems. It stimulates division and differentiation of sympathetic and embryonic sensory neurons.

Subunit structure: Homodimer.

Subcellular location: Secreted.

Involvement in disease: Defects in NGF are the cause of hereditary sensory and autonomic neuropathy type 5 (HSAN5) [

MIM:608654]. The hereditary sensory and autonomic neuropathies are a genetically and clinically heterogeneous group of disorders characterized by degeneration of dorsal root and autonomic ganglion cells, and by sensory and/or autonomic abnormalities. HSAN5 patients manifest loss of pain perception and impaired temperature sensitivity, ulcers, and in some cases self-mutilation. The autonomic involvement is variable. Ref.17
