(Figure. The binding activity of NOS2 with UCHL5.Nitric oxide synthase 2, inducible (NOS2) is a member of Nitric oxide synthases (NOSs) family. Nitric oxide synthases (NOSs) are a family of enzymes catalyzing the production of nitric oxide (NO) from L-arginine. NO is an important cellular signaling molecule. It helps modulate vascular tone, insulin secretion, airway tone, and peristalsis, and is involved in angiogenesis and neural development. Besides, Ubiquitin Carboxyl Terminal Hydrolase L5 (UCHL5) has been identified as an interactor of NOS2, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse NOS2 and recombinant mouse UCHL5. Briefly, NOS2 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to UCHL5-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-NOS2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of NOS2 and UCHL5 was shown in Figure 1, and this effect was in a dose dependent manner.)

The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: mouse thymus tissue lysates,Lane 2: human 22RV1 whole cell lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NFAT2 antigen affinity purified polyclonal antibody at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for NFAT2 at approximately 101KD. It has been reported that IL-6 significantly increased the MMP-10 protein lever in the human lung cancer A549 cells through the JAK/STAT signaling pathway. (Suppressor of cytokine signaling 3 (SOCS3) is a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signaling (SOCS), family that negatively regulates cytokine signal transduction. SOCS3 is feedback inhibitors of the Janus kinase (JAK) and signal transducer and activator of transcription (STAT) signaling pathway. Inhibits cytokine signal transduction by binding to tyrosine kinase receptors including gp130, LIF, erythropoietin, insulin, IL12, GCSF and leptin receptors. SOCS3 also can bind to JAK2 kinase inhibits its kinase activity and nhibits insulin signaling in adipose tissue and the liver. Besides, Glycoprotein 130 (gp130) has been identified as an interactor of SOCS3, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse SOCS3 and recombinant mouse gp130. Briefly, SOCS3 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to SOCS3-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-SOCS3 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of SOCS3 and gp130 was shown in Figure 1, and this effect was in a dose dependent manner.)
