Activity Data (IL7 (Interleukin 7) is a hematopoietic growth factor secreted by stromal cells in the bone marrow and thymus. The interaction between IL17 and the IL7 receptor triggers a cascade of signals important for T-cell development within the thymus and survival within the periphery. It is reported that IL-7 acts on both resting and activated T cells, including Jurkat cells. Thus, a proliferation assay of recombinant human IL7 was conducted using Jurkat cells. Briefly, Jurkat cells were seeded into triplicate wells of 96-well plates at a density of 10, 000 cells/well in RPMI-1640 with the addition of various concentrations of IL7. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Cell proliferation of Jurkat cells after incubation with IL7 for 72h observed by inverted microscope was shown in Figure 1. The CCK-8 data was shown in Figure 2. It was obvious that IL7 significantly promoted cell proliferation of Jurkat cells.)

Activity Data (Figure. The binding activity of PAPPA with Plg.Pregnancy-associated plasma protein A (PAPPA), also known as pappalysin-1, is a secreted protease whose main substrate is insulin-like growth factor binding proteins. PAPPA's proteolytic function is activated upon collagen binding. It is thought to be involved in local proliferative processes such as wound healing and bone remodeling. Low plasma level of this protein has been suggested as a biochemical marker for pregnancies with aneuploid fetuses (fetuses with an abnormal number of chromosomes). Besides, Plasminogen (Plg) has been identified as an interactor of PAPPA, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PAPPA and recombinant human Plg. Briefly, PAPPA were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to Plg-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PAPPA pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of PAPPA and Plg was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Figure. The binding activity of APOA1 with Hpt.Apolipoprotein A1 (APOA1) is the major protein component of HDL particles in plasma. It is a cofactor for lecithin cholesterolacyltransferase (LCAT) which is responsible for the formation of most plasma cholesteryl esters. ApoA1 was also isolated as a prostacyclin (PGI2) stabilizing factor, and thus may have an anticlotting effect. ApoA1 is often used as a biomarker for prediction of cardiovascular diseases. Besides, Haptoglobin (Hpt) has been identified as an interactor of APOA1, thus a binding ELISA assay was conducted to detect the interaction of recombinant human APOA1 and recombinant human Hpt. Briefly, APOA1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to Hpt-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Deoxyribonuclease I (usually called DNase I) is a nonspecific endonuclease that cleaves DNA preferentially at phosphodiester linkages adjacent to a pyrimidine nucleotide, yielding 5'-phosphate-terminated polynucleotides with a free hydroxyl group on position 3', on average producing tetranucleotides. It acts on single-stranded DNA, double-stranded DNA, and chromatin. DNase I can be activated by bivalent metals such as Mg2 and Ca2 . This endonuclease enzyme is common reagents used in biochemical methods requiring diestion of DNA and recovery of RNA, or where DNA is to be removed without affecting structural proteins or enzymes. For example, DNase I is frequently used to remove template DNA following in vitro transcription, and to remove contaminating DNA in total RNA preparations (especially those from transfected cells that may contain plasmid DNA), used for ribonuclease protection assays, cDNA library contraction, and RT-PCR. Activity Data (Nerve growth factor (NGF) is a neurotrophic factor and neuropeptide primarily involved in the regulation of growth, maintenance, proliferation, and survival of certain target neurons. As reported, when the pheochromocytoma cell line PC12 is exposed to nerve growth factor (NGF), the cells respond over a period of a week by ceasing cell division and extending neurites (Greene and Tischler, 1976). The cells were grown in Ham's F12K containing 5% fetal calf serum and 10% horse serum on  polylysine or collagen coated plates. When cells reached log phase growth , fresh medium was added together with 10ng/mL of NGF, then cells were observed by inverted microscope everyday.Cell division ceasing and differentiation of PC12 cells after incubation with NGF (10ng/mL) for 6 days was shown in Figure1.Control group which received no NGF displayed no neurite outgrowth and cells multiply rapidly.)

Activity Data (Mechanism: MMP2 is a zinc-dependent enzymes capable of cleaving components of the extracellular matrix, which belongs to the matrix metalloproteinase (MMP) family. It is a gelatinase A, 72kDa type IV collagenase which can hydrolyze gelatin under certain conditions. Gelatin zymography is mainly used for the detection of the gelatinases, MMP-2 and MMP-9 and It is extremely sensitive because levels of 10pg of MMP-2 can already be detected. Briefly, various concentrations of MMP2 (500ng, 125ng, 63ng, 32ng, 16ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphate-polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP2 would hydrolyze gelatin nearby, which was indicated by the white binds on the gel. In this experiment we use heat-denatured MMP2 protein as negative control, and blood sample as positive control.Result 1: Gelatin hydrolysis by recombinant human MMP2 was shown in figure 1.)
