Principle of the Assay: The LumiAb Human IL-3 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-3 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-3 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-3 or Interleukin-3, alternatively known as Hematopoietic Growth Factor or Mast Cell Growth Factor, is a 152 amino acid cytokine protein capable of supporting the proliferation of a broad range of hematopoietic cell types. The IL-3 monomeric protein is expressed from the IL-3 gene located at locus 5q31.1 on chromosome 5. After initial synthesis of the premature IL-3 protein, it undergoes proteolytic cleavage to remove the 19 residue signal sequence from the 133 residue IL-3 peptide, allowing it to properly fold into a mature protein. It is involved in a variety of cell activities such as cell growth, differentiation and apoptosis. IL-3 has been known to possess neurotrophic activity, and may be associated with neurologic disorders. Typically, granulocyte or macrophage colony-stimulating factors (CSF) are cytokines that act in hematopoiesis by controlling the production, differentiation and function of 2 related white cell populations in the blood, the granulocytes and monocytes-macrophages. This CSF induces granulocytes, macrophages, mast cells, stem cells, erythroid cells, eosinophils and megakaryocytes.

NCBI Description

IL31, which is made principally by activated Th2-type T cells, interacts with a heterodimeric receptor consisting of IL31RA (MIM 609510) and OSMR (MIM 601743) that is constitutively expressed on epithelial cells and keratinocytes. IL31 may be involved in the promotion of allergic skin disorders and in regulating other allergic diseases, such as asthma (Dillon et al., 2004 [PubMed 15184896]).[supplied by OMIM, Mar 2008]
