Principle of the Assay: The LumiAb Human IL-21 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-21 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-21 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-21 or Interleukin-21, also known as Za11, is a 155 amino acid cytokine protein with immunoregulatory activity synthesized from the IL21 gene located at locus 4q26-4q27 on chromosome 4. After initial synthesis, the premature cytokine undergoes proteolytic processing, thus removing its 22 residue signal sequence and allowing the 133 residue IL-21 protein to properly fold and mature. Moreover, IL-21 may promote the transition between innate and adaptive immunity. Expressed in activated CD4+ T-cells but not CD8+ T-cells, B-cells or monocytes, it has been suggested that IL-21 may induce the production of IgG1 and IgG3 in B-cells, may play a role in proliferation and maturation of natural killer (NK) cells in synergy with IL-15, and may regulate proliferation of mature B-and T-cells in response to activating stimuli. In synergy with IL-15 and IL-18, together the three cytokines have been suggested to stimulate IFN-gamma production in T-cells and NK cells. Lastly, it has been proposed that during T-cell mediated immune responses, IL-21 may inhibit dendritic cells (DC) activation and maturation.

NCBI Description

This gene encodes a subunit of the heterodimeric cytokine interleukin 23 (IL23). IL23 is composed of this protein and the p40 subunit of interleukin 12 (IL12B). The receptor of IL23 is formed by the beta 1 subunit of IL12 (IL12RB1) and an IL23 specific subunit, IL23R. Both IL23 and IL12 can activate the transcription activator STAT4, and stimulate the production of interferon-gamma (IFNG). In contrast to IL12, which acts mainly on naive CD4(+) T cells, IL23 preferentially acts on memory CD4(+) T cells. [provided by RefSeq, Jul 2008]

IL23A: Associates with IL12B to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak- Stat signaling cascade, stimulates memory rather than naive T- cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis. Belongs to the IL-6 superfamily.
