Principle of the Assay: The LumiAb Human Galectin-3 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human Galectin-3 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human Galectin-3 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Galectin-3 is a galactose-specific lectin which binds IgE. This lectin may mediate with the alpha-3, beta-1 integrin for the stimulation by CSPG4 of endothelial cells migration. Found together with DMBT1, which is required for terminal differentiation of columnar epithelial cells during early embryogenesis. In the nucleus, it acts as a pre-mRNA splicing factor. Galectin-3 is involved in acute inflammatory responses including neutrophil activation and adhesion, chemoattraction of monocytes macrophages, opsonization of apoptotic neutrophils, and activation of mast cells. It probably forms homo-or heterodimers. Additionally, galectin-3 interacts with DMBT1. This lectin forms a complex with ITGA3, ITGB1 and CSPG4. It also interacts with LGALS3BP, LYPD3, CYHR1 and UACA. It is highly expressed in the colonic epithelium and abundant in activated macrophages.

NCBI Description

This gene encodes a gamma-carboxyglutamic acid (Gla)-containing protein thought to be involved in the stimulation of cell proliferation. This gene is frequently overexpressed in many cancers and has been implicated as an adverse prognostic marker. Elevated protein levels are additionally associated with a variety of disease states, including venous thromboembolic disease, systemic lupus erythematosus, chronic renal failure, and preeclampsia.
