Principle of the Assay: The LumiAb Human IL-31 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-31 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-31 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-31, mainly known as Interleukin-31, is a 164 amino acid cytokine protein encoded by the IL31 gene located at locus 12q24.31 on chromosome 12. After initial synthesis, it is proteolytically processed by proteases to remove the 23 residue signal sequence from the chain and allow for IL-31 to properly fold into a mature protein. IL-31, which is made principally by activated Th2-type T cells, interacts with a heterodimeric receptor consisting of IL-31RA and OSMR that is constitutively expressed on epithelial cells and keratinocytes. Moreover, through the interaction with the IL-31 receptor, IL-31 may serve to activate STAT3 and possibly STAT1 and STAT5. IL-31 may also be involved in the promotion of allergic skin disorders and in regulating other allergic diseases such as asthma. Furthermore, expression of IL-31 is typically detected at low levels in the testes, bone marrow, skeletal muscle, kidney, colon, thymus, small intestine and trachea. Protein synthesis of IL-31 is typically up-regulated in skin homing T-cells of patients with atopic dermatitis (AD), in activated circulating T-cells, and in lesional biopsies of patients with allergic contact dermatitis (ACD).
