Principle of the Assay: The LumiAb Human IL-17A Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-17A concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-17A while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-17A, also known as Interleukin-17A, is a 155 amino acid cytokine encoded by the IL17A gene located at locus 6p12 on chromosome 6. After initial synthesis, the protein is proteolytically cleaved into the 23 residue signal peptide and the 132 residue IL-17A mature protein. In terms of secondary structure, the mature IL-17A cytokine contains 2 alpha helices of 3 to 4 residues each followed by 7 beta strands composed of 3 to 9 residues per strand. IL-17A is a pro-inflammatory cytokine produced by activated T-cells that can regulate the activities of nuclear factor kappa B (NF-kappaB) and mitogen-activated protein (MAP) kinases as well as induce stromal cells to produce other pro-inflammatory and hematopoietic cytokines. The cytokine can also stimulate the expression of the intracellular adhesion molecule-1 (ICAM-1) in fibroblasts, IL-6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). Furthermore, high levels of IL-17A are typically associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis.

NCBI Description

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants.
