Principle of the Assay: The LumiAb Human IL-33 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-33 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-33 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-33 or Interleukin-33, also known as Nuclear Factor from High Endothelial Venules, is a 270 amino acid cytokine encoded by the IL33 gene at locus 9p24.1 on chromosome 9. After initial synthesis, the 111 residue propeptide is cleaved from the actual 159 residue IL-33 chain, thus allowing for the cytokine to properly fold. It is proteolytically converted to a mature form by CASP1 in vitro and calpains in vivo. IL-33 is a member of the IL-1 family that potently drives production of T helper-2 (Th2)-associated cytokines. Moreover, IL-33 is a ligand for IL-33R, an IL-1 family receptor that is selectively expressed on Th2 cells and mast cells. The cytokine binds to and signals through IL-1RL1/ST2 and its stimulation recruits MYD88, IRAK1, IRAK4 and TRAF6, followed by phosphorylation of MAPK3/ERK1 and/or MAPK1/ERK2, MAPK14 and MAPK8. Typically, IL-33 is highly expressed in high endothelial venules found in tonsils, Peyer patches and mesenteric lymph nodes, but almost undetectable in placenta.

NCBI Description

This gene encodes a cytokine that acts as a growth and differentiation factor for both B cells and eosinophils. The encoded cytokine plays a major role in the regulation of eosinophil formation, maturation, recruitment and survival. The increased production of this cytokine may be related to pathogenesis of eosinophil-dependent inflammatory diseases. This cytokine functions by binding to its receptor, which is a heterodimer, whose beta subunit is shared with the receptors for interleukine 3 (IL3) and colony stimulating factor 2 (CSF2/GM-CSF). This gene is located on chromosome 5 within a cytokine gene cluster which includes interleukin 4 (IL4), interleukin 13 (IL13), and CSF2 . This gene, IL4, and IL13 may be regulated coordinately by long-range regulatory elements spread over 120 kilobases on chromosome 5q31. [provided by RefSeq, Jul 2013]
