Activity Data (ENDOG (Endonuclease G) is a mitochondrial enzyme that cleaves DNA at double-stranded (DG)n. (DC)n and at single-stranded (DC)n tracts. This protein also has ribonuclease (RNase) and RNase H activities. It has been reported that EndoG forms complexes with some other proteins, including HSP70 (Heat shock 70 kDa protein). Besides, mouse HSPA1A (Heat shock 70 kDa protein 1A) shares similarities with rat HSP1A1 in amino acid sequence with the identity of 98.44%. Thus, a binding ELISA assay was conducted to detect the association of recombinant rat ENDOG with recombinant mouse HSPA1A. Briefly, ENDOG were diluted serially in PBS with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HSP1A1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ENDOG pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of ENDOG with HSP1A1 was shown in Figure 1 and this effect was in a dose dependent manner.)

Activity Data (Adenine Phosphoribosyltransferase (APRT) is an enzyme involved in the purine nucleotide salvage pathway. It functions as a catalyst in the reaction between adenine and phosphoribosyl pyrophosphate (PRPP) to form AMP. Besides, Vascular Cell Adhesion Molecule 1 (VCAM1) has been identified as an interactor of APRT, thus a binding ELISA assay was conducted to detect the interaction of recombinant human APRT and recombinant human VCAM1. Briefly, APRT were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to VCAM1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-APRT pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. Activity Data (Figure. The binding activity of ACE with ACTb.Angiotensin-converting enzyme ACE, is a central component of the renin-angiotensin system (RAS), which controls blood pressure by regulating the volume of fluids in the body. It converts the hormone angiotensin I to the active vasoconstrictor angiotensin II. Therefore, ACE indirectly increases blood pressure by causing blood vessels to constrict. ACE inhibitors are widely used as pharmaceutical drugs for treatment of cardiovascular diseases. Besides, Actin Beta (ACTb) has been identified as an interactor of ACE, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ACE and recombinant human ACTb. Briefly, ACE were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to ACTb-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (JAK2 (Tyrosine-protein kinase JAK2) is a tyrosine kinase involved in various processes such as cell growth, development, differentiation or histone modifications. JAK2 is considered to associate with some type I receptors, including EPOR (Erythropoietin receptor), therefore participates in cellular signal transduction. Thus a binding ELISA assay was conducted to detect the interaction of recombinant human JAK2 and recombinant human EPOR. Briefly, JAK2 were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL JAK2 were then transferred to EPOR-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Figure. The binding activity of ODC with TK1.Ornithine decarboxylase (ODC) is an enzyme can catalyze the decarboxylation of ornithine (a product of the urea cycle) to form putrescine. The ornithine decarboxylation reaction catalyzed by ornithine decarboxylase is the first and committed step in the synthesis of polyamines, particularly putrescine, spermidine and spermine. Polyamines are important for stabilizing DNA structure, the DNA double strand-break repair pathway and as antioxidants. Therefore, ornithine decarboxylase is an essential enzyme for cell growth, producing the polyamines necessary to stabilize newly synthesized DNA. Lack of ODC causes cell apoptosis in embryonic mice, induced by DNA damage. Besides, Thymidine Kinase 1, Soluble (TK1) has been identified as an interactor of ODC, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ODC and recombinant human TK1. Briefly, ODC were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to TK1-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Matrix Metalloproteinase 13 (MMP13) is a member of the matrix metalloproteinase (MMP) family. MMP13 has been proposed to participate in aggrecan degradation associated with osteoarthritis and cleavage of type II collagen in osteoarthritic cartilage explants and in tumor progression and metastasis. In addition, it can cleave type I, III, IV, IX, X and XIV collagens and fibronectin. MMP13 is likely to play a crucial role in the modulation of extracellular matrix degradation and cell-matrix interactions. Although gelatin zymography is mainly used for the detection of the MMP2 and MMP9, it also can be used for MMP13 dection. Briefly, various concentrations of MMP13 (1000ng, 500ng, 250ng, 125ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphat- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP13 would hydrolyze gelatin nearby, which was indicated by the white bands on the gel. In this experiment we use heat-denatured MMP13 protein as negative control, and blood sample as positive control. Result: Gelatin hydrolysis by recombinant rat MMP13 was shown in figure 1.)

Activity Data (Deoxyribonuclease I (usually called DNase I) is a nonspecific endonuclease that cleaves DNA preferentially at phosphodiester linkages adjacent to a pyrimidine nucleotide, yielding 5'-phosphate-terminated polynucleotides with a free hydroxyl group on position 3', on average producing tetranucleotides. It acts on single-stranded DNA, double-stranded DNA, and chromatin. DNase I can be activated by bivalent metals such as Mg2 and Ca2 . This endonuclease enzyme is common reagents used in biochemical methods requiring diestion of DNA and recovery of RNA, or where DNA is to be removed without affecting structural proteins or enzymes. For example, DNase I is frequently used to remove template DNA following in vitro transcription, and to remove contaminating DNA in total RNA preparations (especially those from transfected cells that may contain plasmid DNA), used for ribonuclease protection assays, cDNA library contraction, and RT-PCR. Besides, Actin Beta (ACTb) has been identified as an interactor of DNase I, thus a binding ELISA assay was conducted to detect the interaction of recombinant human DNase I and recombinant human ACTb. Briefly, DNase I were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to ACTb-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-DNase I pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of DNase I and ACTb was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Figure. The binding activity of TGM1 with SNCa.Transglutaminase 1(TGM1) is an enzyme found in cells that make up the outermost layer of the skin (the epidermis). It can catalyze the addition of an alkyl group from an akylamine to a glutamine residue of a protein, forming an alkylglutamine in the protein. TGM1 is involved in the formation of the cornified cell envelope, which is a structure that surrounds skin cells and helps form a protective barrier between the body and its environment. Besides, Synuclein Alpha (SNCa) has been identified as an interactor of TGM1, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse TGM1 and recombinant mouse SNCa. Briefly, TGM1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to SNCa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TGM1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TGM1 and SNCa was shown in Figure 1, and this effect was in a dose dependent manner.)

Briefly, ODC were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to TK1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ODC pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. Activity Data (Figure. The binding activity of CAPN3 with TTN.Calpain 3 is a calcium-dependent cysteine protease mainly expressed in skeletal muscle. In humans, calpain 3 is encoded by the CAPN3 gene. This gene encodes a muscle-specific member of the calpain large subunit family that specifically binds to titin. Mutations in this gene are associated with limb-girdle muscular dystrophies type 2A. Alternate promoters and alternative splicing result in multiple transcript variants encoding different isoforms and some variants are ubiquitously expressed. Besides, Titin (TTN) has been identified as an interactor of CAPN3, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CAPN3 and recombinant human TTN. Briefly, CAPN3 were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to TTN-coated microtiter wells and incubated for 2h at 37 degree C.
