Activity Data (Annexin A5 (ANXA5) is a cellular protein in the annexin group. In flow cytometry, ANXA5 is commonly used to detect apoptotic cells by its ability to bind to phosphatidylserine, a marker of apoptosis when it is on the outer leaflet of the plasma membrane. ANXA5 has been proposed to play a role in the inhibition of blood coagulation by competing for phosphatidylserine binding sites with prothrombin and also to inhibit the activity of phospholipase A1. Besides, Tumor Protein p53 (TP53) has been identified as an interactor of ANXA5, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat ANXA5 and recombinant rat TP53. Briefly, ANXA5 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to TP53-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ANXA5 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of ANXA5 and TP53 was shown in Figure 1, and this effect was in a dose dependent manner.Figure. Activity Data (Interferon gamma (IFNgamma) is a dimerized soluble cytokine that is the only member of the type II class of interferons. The importance of IFNgamma in the immune system stems in part from its ability to inhibit viral replication directly, and most importantly from its immunostimulatory and immunomodulatory effects. Activity Data (FGF22 (Fibroblast growth factor 22) is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities and are involved in a variety of biological processes including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. A proliferation assay was conducted to detect the bioactivity of recombinant mouse FGF22 using 3T3 cells. Briefly, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of FGF22. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of 3T3 cells after incubation with FGF22 for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant FGF22 for 48h. The result was shown in Figure 2. It was obvious that FGF22 significantly increased cell viability of 3T3 cells.)

The binding activity of FGF2 with CASP1.Basic fibroblast growth factor (FGF2), also known as bFGF, FGF-beta is a member of a large family of structurally related heparin-binding proteins (the FGFs) involved in the regulation of cell proliferation, growth and differentiation. It involved in many biological processes including angiogenesis, embryonic development and wound healing. Additionally, FGF2 is a critical component of human embryonic stem cell culture medium. Besides, Caspase 1 (CASP1) has been identified as an interactor of FGF2, thus a binding ELISA assay was conducted to detect the interaction of recombinant human FGF2 and recombinant human CASP1. Briefly, FGF2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CASP1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-FGF2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of FGF2 and CASP1 was shown in Figure 1, and this effect was in a dose dependent manner.)
