(Figure. The binding activity of OC with FUR.Osteocalcin (OC), also known as bone gamma-carboxyglutamic acid-containing protein (BGLAP), is a noncollagenous protein hormone found in bone and dentin. Osteocalcin is secreted solely by osteoblasts and thought to play a role in the body's metabolic regulation and is pro-osteoblastic, or bone-building, by nature. It is also implicated in bone mineralization and calcium ion homeostasis. Osteocalcin acts as a hormone in the body, causing beta cells in the pancreas to release more insulin, and at the same time directing fat cells to release the hormone adiponectin, which increases sensitivity to insulin. Besides, Furin (FUR) has been identified as an interactor of OC, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse OC and recombinant mouse FUR. Briefly, OC were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100mul were then transferred to FUR-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-OC pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. (Figure. The binding activity of FGF9 with FGFR1.Fibroblast Growth Factor 9 (FGF9) is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. FGF9 was isolated as a secreted factor that exhibits a growth-stimulating effect on cultured glial cells. In nervous system, this protein is produced mainly by neurons and may be important for glial cell development. Besides, Fibroblast Growth Factor Receptor 1 (FGFR1) has been identified as an interactor of FGF6, thus a binding ELISA assay was conducted to detect the interaction of recombinant human FGF9 and recombinant human FGFR1. Briefly, FGF9 were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to FGFR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-FGF9 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of FGF9 and FGFR1 was shown in Figure 1, and this effect was in a dose dependent manner.)

(PTH (Parathyroid hormone) is a hormone secreted by the parathyroid glands that is important in bone remodeling. As reported, osteoblast-like cell lines, such as ROS 17/2.8, UMR106, SaOS, U2OS, MG63, that exhibit PTHR1, respond with increased proliferation to PTH. Rat PTH shares similarities with human PTH in amino acid sequence with the identity of 71.3%. Thus, a proliferation assay of rat recombinant PTH was conducted using U2OS cells. Briefly, U2OS cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of PTH. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of U2OS cells after incubation with PTH for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant PTH for 48h. The result was shown in Figure 2. It was obvious that PTH increased cell viability of U2OS cells.)
