Activity Data (Cluster Of Differentiation 276 (CD276) also known as B7-H3 in human, is a member of the B7/CD28 superfamily of costimulatory molecules serving as an accessory modulator of T-cell response. B7 proteins are immunoglobulin (Ig) superfamily members with extracellular Ig-V-like and Ig-C-like domains and short cytoplasmic domains. Among the family members, they share about 20-40% amino acid (aa) sequence identity. CD276 may play a protective role in tumor cells by inhibiting natural-killer mediated cell lysis as well as a role of marker for detection of neuroblastoma cells. It has also been found to enhance the induction of primary cytotoxic T lymphocytes and stimulate IFN-gamma production. Besides, Sialic acid-binding Ig-like lectin 12 (SIGLEC12) has been identified as an interactor of CD276, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CD276 and recombinant human SIGLEC12. Briefly, CD276 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to SIGLEC12-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CD276 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of CD276 and SIGLEC12 was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. Current data suggest that direct application of IL-6 on breast cancer cells inhibits proliferation in ER-positive (estrogen- receptor- positive) cells through the Jak/Stat3 pathway. To test the inhibitory effect of IL-6 on proliferation of ER-positive MCF-7 cell line, cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of IL-6. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC.The inhibitory effect of IL-6 on cell proliferation of MCF-7 cells observed by inverted microscope and detected by CCK-8 was shown in Figure 1 and Figure 2 respectively (Dose-dependent effect was not detected in this case).)

Activity Data (Interleukin-6 (IL-6), a pro-inflammatory cytokine and an anti-inflammatory myokine, plays important roles in the acute phase reaction, inflammation, hematopoiesis, bone metabolism, and cancer progression. It has been reported that IL-6 significantly increased the MMP-10 protein lever in the human lung cancer A549 cells through the JAK/STAT signaling pathway. Briefly, A549 cells were seeded into 6-well cell culture clusters and allowed to grow to 50-70% confluence, then different concentrations of IL-6 was added. After incubated for 24h, the protein levels of MMP-10 in the cell supernatant were determined by Western blot.Result: MMP-10 protein lever significantly increased in A549 cells due to the stimulation of IL-6, the data was shown in Figure 1.)

Activity Data (Perforin 1 (PRF1) is a pore forming cytolytic protein found in the granules of cytotoxic T lymphocytes (CTLs) and NK cells. Upon degranulation, perforin binds to the target cell's plasma membrane, and oligomerises in a Ca2+ dependent manner to form pores on the target cell. Perforin is thought to act by creating holes in the plasma membrane which triggers an influx of calcium and initiates membrane repair mechanisms. These repair mechanisms bring perforin and granzymes into early endosomes. Besides, Granzyme A (GZMA) has been identified as an interactor of PRF1, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse PRF1 and recombinant mouse GZMA. Briefly, PRF1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to GZMA-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PRF1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. Activity Data (IL2 (Interleukin-2) is a cytokine produced by T-cells in response to antigenic or mitogenic stimulation. IL2 is a type of signaling molecule in the immune system, that is required for both T-cell and B-cell proliferation and other activities crucial to regulation of the immune response. Recombinant human IL2 shares 56% AA sequence identity with mouse IL2, suggesting the exist of cross-species activity. Therefore, in order to detect the bioactivity of rhIL2, a cell proliferation assay has been conducted using CTLL-2 mouse cytotoxic T cells. Briefly, CTLL-2 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with or without the addition of various concentrations of IL2. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). 10uL of CCK-8 solution was added to each well of the plate, the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of CTLL-2 cells after incubation with IL2 for 48h observed by inverted microscope was shown in Figure 1.)

Activity Data (Interleukin-6 (IL-6), a pro-inflammatory cytokine and an anti-inflammatory myokine, plays important roles in the acute phase reaction, inflammation, hematopoiesis, bone metabolism, and cancer progression. It has been reported that IL-6 significantly increased the MMP-10 protein lever in the human lung cancer A549 cells through the JAK/STAT signaling pathway. Briefly, A549 cells were seeded into 6-well cell culture clusters and allowed to grow to 50-70% confluence, then different concentrations of IL-6 was added.
