No cross reactivity with other proteins.
No cross reactivity with other proteins.
Immunohistochemistry (Paraffin-embedded Section): 0.5-1mug/ml

Immunohistochemistry (IHC)
(Figure 4. IHC analysis of CTLA4 using anti-CTLA4 antibody (MBS1750305). CTLA4 was detected in paraffin-embedded section of rat spleen tissue. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-CTLA4 Antibody (MBS1750305) overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen. )

Immunohistochemistry (IHC)
(Figure 3. IHC analysis of CTLA4 using anti-CTLA4 antibody (MBS1750305). CTLA4 was detected in paraffin-embedded section of rat lymphaden tissue. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen. )
Western Blot (WB)
(Figure 1. Western blot analysis of CTLA4 using anti-CTLA4 antibody (MBS1750305). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: mouse NIH3T3 whole Cell lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CTLA4 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CTLA4 at approximately 25KD. The expected band size for CTLA4 is at 25KD. )
Protein Function: Inhibitory receptor acting as a major negative regulator of T-cell responses. The affinity of CTLA4 for its natural B7 family ligands, CD80 and CD86, is considerably stronger than the affinity of their cognate stimulatory coreceptor CD28 (By similarity).
NCBI and Uniprot Product Information
NCBI Description
This gene is a member of the immunoglobulin superfamily, and encodes a protein that functions as a negative regulator of T-cell responses. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Aug 2013]
Uniprot Description
Inhibitory receptor acting as a major negative regulator of T-cell responses. The affinity of CTLA4 for its natural B7 family ligands, CD80 and CD86, is considerably stronger than the affinity of their cognate stimulatory coreceptor CD28.

Product Notes
The CTLA4 ctla4 (Catalog #AAA1750305) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CTLA4 Picoband Antibody reacts with Mouse, Rat No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's CTLA4 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistochemistry (IHC), Western Blot (WB). ELISA: 0.1-0.5mug/ml Immunohistochemistry (Paraffin-embedded Section): 0.5-1mug/ml WB: 0.1-0.5mug/ml. Researchers should empirically determine the suitability of the CTLA4 ctla4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CTLA4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
