Specificity
Recognizes a cluster of proteins between 70-80kDa, identified as tyrosinase. Occasionally a minor band at 55kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. This MAb is a useful marker for melanocytes and melanomas.

NCBI Description

The enzyme encoded by this gene catalyzes the first 2 steps, and at least 1 subsequent step, in the conversion of tyrosine to melanin. The enzyme has both tyrosine hydroxylase and dopa oxidase catalytic activities, and requires copper for function. Mutations in this gene result in oculocutaneous albinism, and nonpathologic polymorphisms result in skin pigmentation variation. The human genome contains a pseudogene similar to the 3' half of this gene. [provided by RefSeq, Oct 2008]

Uniprot Description

TYR: This is a copper-containing oxidase that functions in the formation of pigments such as melanins and other polyphenolic compounds. Catalyzes the rate-limiting conversions of tyrosine to DOPA, DOPA to DOPA-quinone and possibly 5,6-dihydroxyindole to indole-5,6 quinone. Increased expression after UVB irradiation. Belongs to the tyrosinase family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The TYR tyr (Catalog #AAA438285) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Tyrosinase (Melanoma Marker) Mouse Monoclonal Antibody reacts with Human. Others not known. and may cross-react with other species as described in the data sheet. AAA Biotech's Tyrosinase can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC) Formalin. Flow Cytometry (0.5-1ug/million cells ) Immunofluorescence (1-2ug/ml) Immunohistology (Formalin-fixed) (0.5-1ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA, pH 7.5-8.5,for 10-20 ming followed by cooling at RT for 20 min). Optimal dilution for a specific application should be determined. Researchers should empirically determine the suitability of the TYR tyr for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Tyrosinase, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
