(IL1alpha (Interleukin-1 alpha) is a member of the interleukin 1 cytokine family. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces cell apoptosis. It is reported that exposure of MCF-7 cells to certain concentration of IL1alpha results in inhibition of cell growth. Thus, an cell proliferation assay of MCF-7 was conducted with the addition of IL1alpha. MCF-7 cells were seeded overnight at a density of 5,000 cells/well, and then treated with or without various concentrations of IL1alpha for 72h, then cells were observed by inverted microscope and cell viability was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours in at 37 degree C.Inhibition of MCF-7 cell proliferation after incubation with IL1alpha for 72h observed by inverted microscope was shown in Figure 1.)

(Interleukin-6 (IL-6), a pro-inflammatory cytokine and an anti-inflammatory myokine, plays important roles in the acute phase reaction, inflammation, hematopoiesis, bone metabolism, and cancer progression. (Growth Regulated Oncogene Beta (GROb) is a small cytokine belonging to the CXC chemokine family which produced by activated monocytes and neutrophils and expressed at sites of inflammation. Hematoregulatory chemokine, which, in vitro, suppresses hematopoietic progenitor cell proliferation. Besides, Dipeptidyl Peptidase IV (DPP4) has been identified as an interactor of GROb, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat GROb and recombinant rat DPP4. Briefly, GROb were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100ul were then transferred to DPP4-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GROb pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of GROb and DPP4 was shown in Figure 1, and this effect was in a dose dependent manner.)

(Suppressor of cytokine signaling 3 (SOCS3) is a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signaling (SOCS), family that negatively regulates cytokine signal transduction. SOCS3 is feedback inhibitors of the Janus kinase (JAK) and signal transducer and activator of transcription (STAT) signaling pathway. Inhibits cytokine signal transduction by binding to tyrosine kinase receptors including gp130, LIF, erythropoietin, insulin, IL12, GCSF and leptin receptors. SOCS3 also can bind to JAK2 kinase inhibits its kinase activity and nhibits insulin signaling in adipose tissue and the liver. (IL-1beta (Interleukin-1 beta) is a proinflammatory and immunoregulatory cytokine involved in a variety of cellular activities. It is produced by activated macrophages as a proprotein, and then proteolytically processed to an active form. It has been reported that IL-1beta-induced IL-6 production is mediated by both PI3K and IRAK4 in A549 cells. To detect the bioactivity of IL-1beta, A549 cells were seeded into 24-well plate at a density of 1x10^5cells/mL, and allowed to attach overnight before treated with or without certain concentrations (1ng/mL, 10ng/mL) of IL1-beta for 4h and IL-6 levels in the cell supernatant were determined by ELISA.IL-6 levels in the cell supernatant of A549 cells increased significantly after stimulated with IL1-beta, the data was shown in Table 1 and Figure 1.)

(Vascular endothelial growth factor C (VEGFC) is a protein that is a member of the platelet-derived growth factor/vascular endothelial growth factor (PDGF/VEGF) family. It plays key roles in the physiology and pathology of many aspects of the cardiovascular system, including vasculogenesis, hematopoiesis, angiogenesis and vascular permeability. To test the effect of VEGFC on cell proliferation of ECV304 endothelium cell line, cells were seeded into triplicate wells of 96-well plates at a density of 2,000cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of VEGFC. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. The dose-effect curve of VEGFC was shown in Figure 1. It was obvious that VEGFC significantly promoted cell proliferation of ECV304 cells. The ED50 for this effect is typically 0.304 to 1.339ug/mL.)

The binding activity of GM-CSF with CSF2Ra.Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2), is a monomeric glycoprotein secreted by macrophages, T cells, mast cells, natural killer cells, endothelial cells and fibroblasts that functions as a cytokine. GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils, and basophils) and monocytes. It also has some effects on mature cells of the immune system. These include, for example, inhibiting neutrophil migration and causing an alteration of the receptors expressed on the cells surface. GM-CSF signals via signal transducer and activator of transcription, STAT5. In macrophages, it has also been shown to signal via STAT3. Besides, Colony Stimulating Factor 2 Receptor Alpha (CSF2Ra) has been identified as an interactor of GM-CSF, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat GM-CSF and recombinant rat CSF2Ra. Briefly, GM-CSF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CSF2Ra-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GM-CSF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of GM-CSF and CSF2Ra was shown in Figure 1, and this effect was in a dose dependent manner.)

(IFN-gamma is a dimerized soluble cytokine that is the only member of the type II class of interferons. The importance of IFNgamma in the immune system stems in part from its ability to inhibit viral replication directly, and most importantly from its immunostimulatory and immunomodulatory effects. As reported, IFNgamma is an important activator of human monocytic THP1 cells. Therefore, THP-1 cells were incubated in RPMI 1640 with various concentration of IFN-gamma, then cells were observed by inverted microscope everyday. After stimulated with IFN-gamma (2ng/mL) for 5 days, morphological changes occurred in THP1 cells which displayed the shape of fusiform or polygon and were more likely to adhere.
