Solubility
It is recommended to reconstitute the lyophilized GRO-beta Rat in sterile 18M Omega -cm H2O not less than 100 ug/ml, which can then be further diluted to other aqueous solutions.
Biological Activity
The biological activity was determined by its ability to chemoattract total human neutrophils using a concentration range of 1.0-10.0 ng/ml, corresponding to a specific activity of 100,000-1,000,000 units/mg.
Preparation and Storage
Lyophilized GRO-b although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution CXCL2 should be stored at 4 degree C between 2-7 days and for future use below -18 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.
Related Product Information for rGRO b active protein
Description: GRO-Beta Rat Recombinant also called Rat MIP-2 produced in E Coli is a single, non-glycosylated, polypeptide chain containing 73 amino acids and having a molecular mass of 7923 Dalton. The GRO-b is purified by proprietary chromatographic techniques.

Introduction: Chemokine (C-X-C motif) ligand 2 (CXCL2) is a small cytokine belonging to the CXC chemokine family that is also called macrophage inflammatory protein 2-alpha (MIP2-alpha), Growth-regulated protein beta (Gro-beta) and Gro oncogene-2 (Gro-2). CXCL2 is 90% identical in amino acid sequence as a related chemokine, CXCL1. This chemokine is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. The gene for CXCL2 is located on human chromosome 4 in a cluster of other CXC chemokines. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2.

NCBI Description

chemokine involved in the pulmonary inflammatory response [RGD, Feb 2006]

Uniprot Description

CXCL3: Ligand for CXCR2. Has chemotactic activity for neutrophils. May play a role in inflammation and exert its effects on endothelial cells in an autocrine fashion. In vitro, the processed form GRO-gamma(5-73) shows a fivefold higher chemotactic activity for neutrophilic granulocytes. Belongs to the intercrine alpha (chemokine CxC) family.

Protein type: Secreted; Motility/polarity/chemotaxis; Secreted, signal peptide

Cellular Component: extracellular space

Molecular Function: chemokine activity; CXCR chemokine receptor binding

Biological Process: neutrophil chemotaxis; wound healing; leukocyte chemotaxis; response to glucocorticoid stimulus; response to amphetamine; positive regulation of leukocyte chemotaxis; response to estradiol stimulus; regulation of cell proliferation; G-protein coupled receptor protein signaling pathway; elevation of cytosolic calcium ion concentration; response to heat; response to molecule of bacterial origin; response to gamma radiation; immune response; inflammatory response

Product Notes

The rGRO b cxcl2 (Catalog #AAA144089) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: VVVASELRCQ CLTTLPRVDF KNIQSLTVTP PGPHCAQTEV IATLKDGHEV CLNPEAPLVQ RIVQKILNKG KAN. It is sometimes possible for the material contained within the vial of "GRO-beta/MIP-2, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
