Activity Data (Transforming growth factor alpha (TGF-alpha), a ligand for the epidermal growth factor receptor, which activates a signaling pathway for cell proliferation, differentiation and development. To test the effect of TGF-alpha on cell proliferation of 3T3 fibroblasts, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2, 000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of TGF-alpha. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37 degree C.Cell proliferation of 3T3 cells after incubation with TGF-alpha for 72h observed by inverted microscope was shown in Figure 1.)

Activity Data (Protein Kinase R (PKR) is activated by double-stranded RNA (dsRNA), the synthesis of which is caused virally. PKR can also be activated by the protein PACT or by heparin. It plays a key role in the innate immune response to viral infection and is also involved in the regulation of signal transduction, apoptosis, cell proliferation and differentiation. Besides, Cyclin Dependent Kinase 1 (CDK1) has been identified as an interactor of PKR, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PKR and recombinant human CDK1. Briefly, PKR were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CDK1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PKR pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. The EGFR is essential for ductal development of the mammary glands, and agonists of the EGFR such as amphiregulin, TGF-alpha, and heregulin induce both ductal and lobuloalveolar development even in the absence of estrogen and progesterone. Besides, Actin Beta (ACTb) has been identified as an interactor of EGFR2, thus a binding ELISA assay was conducted to detect the interaction of recombinant human EGFR2 and recombinant human ACTb. Activity Data (IDO (Indoleamine 2,3-dioxygenase 1) is a heme enzyme that catalyzes the first and rate-limiting step in tryptophan catabolism to N-formyl-kynurenine. This enzyme acts on multiple tryptophan substrates including D-tryptophan, L-tryptophan, 5-hydroxy-tryptophan, tryptamine, and serotonin. Activity Data (Figure. Cell proliferation of HCT116 cells after stimulated with PK2.Prokineticin 2 (PK2) is a memeber of prokineticin family. Prokineticin is a secreted protein that potently contracts gastrointestinal smooth muscle. They are thought to be involved in several important physiological processes like neurogenesis, tissue development, angiogenesis, and nociception. Other important physiological roles the Bv8/Prokineticins (PKs) are involved in may include cancer, reproduction, and regulating physiological functions that influence circadian rhythms like hormone secretion, ingestive behaviors, and the sleep/wake cycle. To test the effect of PK2 on cell proliferation, HCT116 colon cancer cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM containing various concentrations of recombinant human PK2. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of HCT116 cells after incubation with PK2 for 96h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant PK2 for 96h. The result was shown in Figure 2.
