Source Note
Optimized DNA sequence encoding Human extracellular domain of human CD16a including a C-terminal His tag was expressed in HEK293 cells.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Biological Activity
Recombinant CD16a activity was measured by the ability to bind human IgG with an affinity constant of 180nM.
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL.
Molecular Weight Note
Recombinant CD16a is a monomer protein consisting of 203 amino acid residue subunits, due to glycosylation migrates as an approximately 50 kDa protein on SDS-PAGE.
Preparation and Storage
Recombinant CD16a, as supplied, can be stored in working aliquots at 2-8°C for one month, or at -20°C to -70°C for twelve months.

Avoid repeated freeze/thaw cycles.

Human CD16 is encoded by two genes, FcγRIIIA and FcγRIIIB. FcγRIIIa is a transmembranous isoform of the receptor expressed on the surface of natural killer (NK) cells, macrophages, a subset of monocytes and of T cells in association with dimers of the γ-chain of FcεRI and/or ζ-chain of T-cell receptors. FcγRIIIb is a glycosylphosphatidylinositol -anchored isoform restricted to neutrophils. FcγRIII plays a significant role in the clearance of immune complexes, antibody- dependent cellular cytotoxicity of K/NK cells,phagocytosis and antigen presentation.

NCBI Description

This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other other antibody-dependent responses. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
