Principle of the Assay: The LumiAb Human beta-NGF Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human beta-NGF concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human beta-NGF while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human beta-NGF, also known as Nerve Growth Factor beta, is a 241 amino acid cytokine protein encoded by the NGFB gene at locus 1p13.1 on chromosome 1. After initial synthesis of the premature beta-NGF peptide, it is subject to proteolytic processing and cleavage, thus separating the 18 residue signal sequence, the 103 residue propeptide and the 120 residue mature beta-NGF protein. beta-NGF is a secreted protein with sympathetic as well as embryonic nerve growth stimulating activity which homodimerizes and is incorporated into a larger complex that is involved in the regulation of growth and differentiation of sympathetic and certain sensory neurons. Mutations in this gene have often been associated with hereditary sensory and autonomic neuropathy type 5 (HSAN5) and dysregulation of the NGFB's expression can lead to allergic rhinitis.

NCBI Description

This gene encodes a secreted ligand of the TGF-beta (transforming growth factor-beta) superfamily of proteins. Ligands of this family bind various TGF-beta receptors leading to recruitment and activation of SMAD family transcription factors that regulate gene expression. The encoded preproprotein is proteolytically processed to generate each subunit of the disulfide-linked homodimer, which plays a role in bone and cartilage development. Duplication of a regulatory region downstream of this gene causes a form of brachydactyly characterized by a malformed index finger and second toe in human patients. [provided by RefSeq, Jul 2016]

Induces cartilage and bone formation (PubMed:3201241). Stimulates the differentiation of myoblasts into osteoblasts via the EIF2AK3-EIF2A- ATF4 pathway. BMP2 activation of EIF2AK3 stimulates phosphorylation of EIF2A which leads to increased expression of ATF4 which plays a central role in osteoblast differentiation. In addition stimulates TMEM119, which upregulates the expression of ATF4 (PubMed:24362451).
