Principle of the Assay: The LumiAb Murine SCF Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Murine SCF concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Murine SCF while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Murine SCF or Stem Cell Factor, also known as Kit Ligand, is a 273 amino acid cytokine protein expressed from the Kitl gene located on chromosome 10. After initial synthesis and translocation, the 25 residue signal sequence is cleaved, allowing for the 248 residue SCF to properly fold and mature. If the SCF or Kit Ligand is cleaved again on the C-terminal end at residue 190, then the soluble Kit Ligand is formed. SCF, a non-covalently linked homodimer, is known to stimulate the proliferation of mast cells and is able to augment the proliferation of both myeloid and lymphoid hematopoietic progenitors in bone marrow culture. Also, SCF serves to mediate cell-cell adhesion and acts synergistically with other cytokines and interleukins.

Uniprot Description

TNFRSF11A: Receptor for TNFSF11/RANKL/TRANCE/OPGL; essential for RANKL-mediated osteoclastogenesis. Involved in the regulation of interactions between T-cells and dendritic cells. Binds to the clefts between the subunits of the TNFSF11 ligand trimer to form a heterohexamer. Interacts with TRAF1, TRAF2, TRAF3, TRAF5 and TRAF6. Interacts (via cytoplasmic domain) with GAB2. Ubiquitous expression with high levels in skeletal muscle, thymus, liver, colon, small intestine and adrenal gland.

Product Notes

The Mouse TNFRSF11a tnfrsf11a (Catalog #AAA9510854) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9510854 CLIA Kit recognizes Mouse TNFRSF11a. It is sometimes possible for the material contained within the vial of "sRANK Ligand, CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
