Principle of the Assay: The LumiAb Human P1GF Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human P1GF concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human P1GF while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human P1GF-1, also known as Placenta Growth Factor, is a 221 amino acid growth factor protein encoded by the PGF gene located at locus 14q24.3 on chromosome 14. After initial synthesis, the 18 residue signal sequence is removed, allowing for the proper folding and maturation of the 203 residue PLGF-1 protein. PLGF-1 is essentially a growth factor active in angiogenesis and endothelial cell growth, stimulating their proliferation and migration. It binds to the receptor FLT1/VEGFR-1 while isoform PlGF-2 binds NRP1/neuropilin-1 and NRP2/neuropilin-2 in a heparin-dependent manner. The subunit structure is known to be an antiparallel homodimer connected via disulfide linkages, but also can be found as a heterodimer with VEGFA/VEGF. Meanwhile, the PLGF-3 isoform can be found as both as a homodimer and as a monomer. While the three isoforms are present in most placental tissues, PLGF-2 is specific to early (8 week) placenta and only PLGF-1 is found in the colon and mammary carcinomas.

NCBI Description

This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2009]
