Testing Data (Figure 1. Response of ACTOne GPBAR1 (TGR5) cell line & parental cell line to LCA. ACTOne GPBAR1 cells and parental cells were plated overnight in 20 ul culture medium on a BD Biocoat 384 well plate. Human GPBAR1 ACTOne is a HEK293-CNG cell line that expresses recombinant human GPBAR1. HEK293-CNG cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization which can be easily measured with fluorescent Membrane Potential Dye (Cat# MBS430187). The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR, or a fluorescence microplate reader.

NCBI Description

This gene encodes a member of the G protein-coupled receptor (GPCR) superfamily. This enzyme functions as a cell surface receptor for bile acids. Treatment of cells expressing this GPCR with bile acids induces the production of intracellular cAMP, activation of a MAP kinase signaling pathway, and internalization of the receptor. The receptor is implicated in the suppression of macrophage functions and regulation of energy homeostasis by bile acids. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq, Jul 2008]
