Short Description: Beta-Glucosyltransferase, also called BGT, is a DNA-modifying enzyme. It is encoded by a number of bacteriophages belonging to the T-even group. It catalyzes the transfer of glucose (Glc) from uridine diphosphoglucose (UDP-Glc) to 5-hydroxymethylcytosine (5-HMC) in double-stranded DNA. BGT shows little DNA sequence specificity but modify only 5-HMC bases. The glucosylation protects the infecting viral DNA from host nucleases. In addition to its protective role, the glucosylation has also been implicated in the control of phage-specific gene expression by influencing transcription in vivo and in vitro.

Uniprot Description

Catalyzes the transfer of glucose from uridine diphosphoglucose to 5-hydroxymethyl cytosine of T4 DNA to yield glucosyl 5-hydroxymethyl cytosine (glc-HMC) (PubMed:22229759). This DNA process seems to occur immediately after DNA synthesis since the DNA alpha-glucosyltransferase interacts with the clamp protein gp45 (PubMed:22229759). The glc-HMC modification protects the phage genome against its own nucleases and the host restriction endonuclease system (PubMed:22229759). The glc-HMC modification also protects against the host CRISPR-Cas9 defense system (PubMed:26081634).
