Members of this calcium-dependent phospholipid-binding protein family play a role in the regulation of cellular growth and in signal transduction pathways. This protein functions as an autocrine factor which heightens osteoclast formation and bone resorption. It is a calcium-dependent phospholipid-binding protein whose function is to help organize exocytosis of intracellular proteins to the extracellular domain. Besides, S100 Calcium Binding Protein A10 (S100A10) has been identified as an interactor of ANXA2, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse ANXA2 and recombinant mouse S100A10. Briefly, ANXA2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to S100A10-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ANXA2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. AMH signals through a characteristic receptor consisting of a type I and a type II receptor serine/threonine kinase. Especially the type II receptor is unique and specific receptor for AMH. Besides, Mothers Against Decapentaplegic Homolog 9 (Smad9) has been identified as an interactor of AMH, thus a binding ELISA assay was conducted to detect the interaction of recombinant human AMH and recombinant human (Smad9) Briefly, AMH were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to Smad9-coated microtiter wells and incubated for 2h at 37 degree C. (Glucose and other reducing sugars can react non-enzymatically with the amino groups of proteins to form compounds called advanced glycation end products (AGEs). AGEs exert their cellular functions via the interaction with receptor for advanced glycation end products (RAGE). It has been reported that AGE stimulates the differentiation and proliferation of 3T3, thus a proliferation assay was conducted using 3T3 cells. Briefly, 3T3 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of AGE. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of 3T3 cells after incubation with AGE for 48h observed by inverted microscope was shown in Figure1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with recombinant AGE for 48h. The result was shown in Figure 2. It was obvious that AGE significantly increased cell viability of 3T3 cells.)
