Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FASN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FASN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FASN is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FASN bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The enzyme encoded by this gene is a multifunctional protein. Its main function is to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha. [provided by RefSeq, Jul 2008]
