(Figure 1. Western blot analysis of ABCB4 using anti-ABCB4 antibody (MBS177897).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: MCF-7 Whole Cell Lysate,Lane 2: SW620 Whole Cell Lysate,Lane 3: 22RV1 Whole Cell Lysate,Lane 4: SKOV Whole Cell Lysate.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ABCB4 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for ABCB4 at approximately 142KD. The expected band size for ABCB4 is at 142KD. )

The binding activity of EGRF2 with ACTb.The epidermal growth factor receptor (EGFR; ErbB-1; HER1 in humans) is a transmembrane protein that is a receptor for members of the epidermal growth factor family (EGF family) of extracellular protein ligands. Epidermal growth factor receptor 2 (EGFR2) is a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. The EGFR is essential for ductal development of the mammary glands, and agonists of the EGFR such as amphiregulin, TGF-alpha, and heregulin induce both ductal and lobuloalveolar development even in the absence of estrogen and progesterone. Besides, Actin Beta (ACTb) has been identified as an interactor of EGFR2, thus a binding ELISA assay was conducted to detect the interaction of recombinant human EGFR2 and recombinant human ACTb. Briefly, EGFR2 were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to ACTb-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-EGFR2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of EGFR2 and ACTb was shown in Figure 1, and this effect was in a dose dependent manner.)

Thioflavin T is a fluorescent dye that binds to beta sheet-rich structures, such as those in alpha Synuclein aggregates. Upon binding, the emission spectrum of the dye experiences a red-shift, and increased fluorescence intensity. Thioflavin T emission curves show increased fluorescence (correlated to alpha Synuclein protein aggregation) over time when 10 nM of active alpha Synuclein aggregate (SPR-324) is combined with 100 uM of active alpha Synuclein monomer (SPR-323), as compared to active alpha Synuclein aggregate (SPR-324) and active alpha Synuclein monomer (SPR-323) alone.
