Activity Data (Osteopontin (OPN), a multifunctional phosphorylated glycoprotein, plays an important role in neutrophil recruitment and was found to induce the expression of proinflammatory chemokines including MCP-1 and MIP-1beta which promote migration and recruitment of inflammatory cells. It has been reported that OPN induces MCP-1 expression through the NF-kappa B pathways in MCF-7 breast cancer cell line. Briefly, MCF-7 cells were seeded overnight at a density of 1x10^5 cells/mL, and treated with or without various concentrations of OPN for 24h and MCP-1 levels in the cell supernatant were determined by ELISA.MCP-1 levels in the cell supernatant of MCF-7 cells increased significantly after stimulated with OPN, the data was shown in Table 1 and Figure 1.)

Western blot analysis of PF4 using anti-PF4 antibody (MBS1750500).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: rat spleen tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PF4 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for PF4 at approximately 11KD. Designation of population shown within each dot-plot is indicated above the dot-plot. Leukocytes were identified as viable (a) non-doublet (b) cells with typical light scatter properties of leukocytes (c). Then, macrophages were gated simply as CD203ahi leukocytes (d) and marked with blue color. Monocytes were gated as CD203alow/- SWC8-(e) CD172ahi(f) leukocytes where the CD203alow/- region was defined as the complementary region to the CD203ahi region. Then, SLA-DR+ monocytes were marked with red color and SLA-DR- monocytes were marked with green color (g). SLA-DR- region was defined as the complementary region to the SLA-DR+ region. Gating order is shown in the scheme (h). Testing Data (Published customer image: Confocal neutrophil-cytokine double labeling experiments using incised skin. For these studies skin from the wound edges was harvested 2 hours after incision. After sectioning, the tissue was exposed to anti-neutrophil and anti-IL-1beta antibodies followed by the application of CY3 (green fluorescence, neutrophils, panel A) and FITC (red, IL-1beta, panel B) conjugated secondary antibodies. Panel C presents the merged image with arrows pointing to several strongly double labeling cells. The scale bar in panel C is 50 um in length. These micrographs were taken of dermal tissue under 630x magnification. From: Clark JD, Shi X, Li X, Qiao Y, Liang D, Angst MS, Yeomans DC. Morphine reduces local cytokine expression and neutrophil infiltration after incision. Mol Pain. 2007 Oct 2;3:28.)
