Western Blot (WB) (Figure 1. Western blot analysis of Thrombopoietin using anti-Thrombopoietin antibody (MBS1750819).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: mouse liver tissue lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. Activity Data (The interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. Upon activation by IL4, Th2 cells subsequently produce additional IL4 in a positive feedback loop. IL4 has many biological roles, including the stimulation of activated B-cell and T-cell proliferation, and the differentiation of B cells into plasma cells. It is a key regulator in humoral and adaptive immunity. IL4 induces B-cell class switching to IgE, and up-regulates MHC class II production. IL4 decreases the production of Th1 cells, macrophages, IFN-gamma, and dendritic cell IL12. Besides, Interleukin 2 Receptor Gamma (IL2Rg) has been identified as an interactor of IL4, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse IL4 and recombinant mouse IL2Rg. Activity Data (Figure. Cell proliferation of A549 cells after stimulated with GREM1.Gremlin (GREM1) is an inhibitor in the TGF beta signaling pathway. GREM1 and other BMP antagonists are important in the survival of cancer stroma survival and proliferation in some cancers. The protein expression is found in many cancers and is thought to play important roles in uterine cervix, lung, ovary, kidney, breast, colon, pancreas, and sarcoma carcinomas. A proliferation assay was conducted to detect the bioactivity of recombinant mouse GREM1 using A549 cells. Briefly, A549 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM prior to the addition of various concentrations of GREM1. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly,10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of A549 cells after incubation with GREM1 for 96h observed by inverted microscope was shown in Figure1. Activity Data (IL17C (Interleukin-17C) is a T cell-derived cytokine that shares the sequence similarity with IL17. IL17C is thought to play a crucial role in innate immunity of the epithelium. IL17C binds with affinity to IL17RA. Thus, a binding ELISA assay was constructed to detect the association of rhIL17C with IL17RA. Activity Data (Interleukin-17 receptor A (IL17RA) is a cytokine receptor which binds interleukin 17A (IL17A). IL17A is a proinflammatory cytokine secreted by activated T-lymphocytes.
