(IL17C (Interleukin-17C) is a T cell-derived cytokine that shares the sequence similarity with IL17. IL17C is thought to play a crucial role in innate immunity of the epithelium. IL17C binds with affinity to IL17RA. Thus, a binding ELISA assay was constructed to detect the association of rhIL17C with IL17RA. Briefly, rhIL17C were diluted serially in PBS with 0.1% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17RA-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately.
