(issue: Sf-1:Cre mice crossed to the Z/EG reporter line. Mouse brain (coronal view, 20X magnification). Fixation: 4%PFA/PBS with o/n fixation, and subsequently transferred to a 30% sucrose solution. Antigen retrieval: frozen in OCT freezing medium (Sakura) and cryostat sectioned at 40 microns. Goat anti-GFP was used at 1:500 dilution in free floating imunnohistochemistry to detect GFP. Fluorchrome conjugated Anti-goat IgG secondary antibody was used for detection at 1:500 at 1:10,000 for 45 min at RT. Besides, Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) has been identified as an interactor of TK1, thus a binding ELISA assay was conducted to detect the interaction of recombinant human TK1 and recombinant human GAPDH. Briefly, TK1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to GAPDH-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TK1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TK1 and GAPDH was shown in Figure 1, and this effect was in a dose dependent manner.)
