( Western Blot using anti-CD4 antibody CE9.1 (Clenoliximab) Human thyroid tumour samples were resolved on a 15% SDS PAGE gel and blots probed with the human IgG4-chimeric version of CE9.1 at 1.5 ug/ml before detection using an anti-human IgG4 secondary antibody. Protein was detected by chemiluminescence. The expected unmodified running size for CD4 is 51kDa (arrowhead). Here some non-specific bands can also be seen. successfully detected CD4 in human thyroid tissue samples.)

This gene encodes a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus. This gene is expressed not only in T lymphocytes, but also in B cells, macrophages, and granulocytes. It is also expressed in specific regions of the brain. The protein functions to initiate or augment the early phase of T-cell activation, and may function as an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system. Multiple alternatively spliced transcript variants encoding different isoforms have been identified in this gene. [provided by RefSeq, Aug 2010]

The Anti-CD4 [CE9.1 (Clenoliximab)] reacts with Human, Chimpanzee and may cross-react with other species as described in the data sheet. AAA Biotech's CD4 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Flow Cytometry (FC/FACS), Block, SPR, Immunohistochemistry (IHC). The antibody binds specifically to CD4, a molecule which is part of the immunolgobulin super-family, expressed on the surface of helper T cells. The CD4 molecule acts as a co-receptor in the activation of T cells, aiding the interaction between the TCR and MHC II of the antigen-presenting cell. The antibody is classified as "primatized", as the variable regions of the light and heavy chains are obtained from primates, while the lamda light chain and gamma4 heavy chain constant domains are human. The antibody was engineered from the primatized IgG1 monoclonal antibody Keliximab by substituting two key residues. The role of the antibody as an immunosuppressor has been tested in vivo, by intravenous administration in chimpanzees and human volunteers. The antibody reached Phase II clinical trials with Biogen-Idec. Researchers should empirically determine the suitability of the CD4 cd4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
