Species Reactivity Note
The antibody has been directly tested for reactivity in Western blots with Xenopus tissue. It is anticipated that the antibody will react with human, mouse, rat and zebra fish based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance
Considerable evidence indicates that a polo-like kinase (PLK) plays an important role in cell cycle regulation. PLK is also required for bipolar spindle formation, activation of the anaphase-promoting complex/cyclosome, and cytokinesis. Recent work led to the identification of a PLKK that is thought to be responsible for activation of PLK. Recent work (Erikson, et al., 2004) has shown that PLKK is in turn activated by phosphorylation at three sites (Ser482, Ser486 and Ser490). Thus activation of PLK is thought to involve a kinase cascade involving the phosphorylation of Ser482,486,490 in PLKK.
Preparation and Storage
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.

Uniprot Description

Function: Serine/threonine-protein kinase that performs several important functions throughout M phase of the cell cycle, including the regulation of centrosome maturation and spindle assembly, the removal of cohesins from chromosome arms, the inactivation of anaphase-promoting complex/cyclosome (APC/C) inhibitors, and the regulation of mitotic exit and cytokinesis. Polo-like kinase proteins acts by binding and phosphorylating proteins are that already phosphorylated on a specific motif recognized by the POLO box domains. Phosphorylates cdc25, pkmyt1/myt1, stag2/sa2, tpx2. Plays multiple essential roles during mitosis. Phosphorylates the N-terminal domain of cdc25, which leads to cyclin b-cdc2 activation and mitotic entry. Also required for organization of bipolar spindles, and for exit from mitosis. Involved in kinetochore functions and sister chromatid cohesion by phosphorylating stag2/sa2. Ref.1 Ref.3 Ref.4 Ref.5 Ref.6 Ref.7 Ref.8 Ref.9 Ref.10

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Enzyme regulation: Activated by phosphorylation of Thr-201. Ref.4

Subunit structure: Interacts with plk1 and kif2a. Ref.9 Ref.10

Subcellular location: Nucleus. Cytoplasm › cytoskeleton › microtubule organizing center › centrosome. Cytoplasm › cytoskeleton › spindle. Midbody

By similarity. Note: Localized at centrosomes at prophase, spindles at metaphase, and at the midbody during cytokinesis. Ref.4

Induction: By growth-stimulating agents

By similarity. Ref.4

Domain: The POLO box domains act as phosphopeptide-binding module that recognize and bind serine-[phosphothreonine/phosphoserine]-(proline/X) motifs. plk1 recognizes and binds docking proteins that are already phosphorylated on these motifs, and then phosphorylates them

By similarity.

Post-translational modification: Activated by phosphorylation on Thr-201 during M phase. Phosphorylated by stk10, leading to activation during oocyte maturation. Ref.3 Ref.4 Ref.5Ubiquitinated by the anaphase promoting complex/cyclosome (APC/C) in anaphase and following DNA damage, leading to its degradation by the proteasome. Protein levels are down-regulated by proteasomal degradation in anaphase

By similarity.

Sequence similarities: Belongs to the protein kinase superfamily. Ser/Thr protein kinase family.Contains 2 POLO box domains.Contains 1 protein kinase domain.

Product Notes

The PLK1 plk1 (Catalog #AAA502103) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Phospho-Ser482,486,490 Polo-Like Kinase Kinase reacts with Xenopus and may cross-react with other species as described in the data sheet. AAA Biotech's Polo-Like Kinase Kinase (Ser482,486,490) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Quality Control: Western blots performed on each lot. WB: 1:1000. Researchers should empirically determine the suitability of the PLK1 plk1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Polo-Like Kinase Kinase (Ser482,486,490), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
